Under certain environmental conditions, it has massive outbreaks, resulting in attacks of healthy trees, becoming a forest pest. It has been proposed that the bark beetle’s microbiome plays a key role in the insect’s ecology, providing nutrients, inhibiting pathogens, and degrading tree defense compounds, among other probable traits. During a study of bacterial associates from I. typographus, we isolated three strains identified as Pseudomonas from different beetle life stages. In this work, we aimed to reveal the taxonomic status of these bacterial strains and to sequence and annotate their genomes to mine possible traits related to a role within the bark beetle holobiont. -

The corresponding gene was cloned by polymerase chain reaction (PCR) and the dimeric enzyme with C-terminal 6-histidines was successfully overproduced and purified from recombinant Escherichia coli. BHCD showed the highest activity against β-CD at pH 7.0 and 50oC. Due to its versatile hydrolysis and transglycosylation activities, BHCD has been confirmed as a member of CDases. However, BHCD can be distinguished from other typical CDases on the basis of its novel multisubstrate specificity. While typical CDases have over 10 times higher activity on β-CD than starch or pullulan, the CD-hydrolyzing activity of BHCD is only 2.3 times higher than pullulan. -

The use of pullulanase (EC 3.2.1.41) has recently been the subject of increased applications in starch-based industries especially those aimed for glucose production. Pullulanase, an important debranching enzyme, has been widely utilised to hydrolyse the α-1,6 glucosidic linkages in starch, amylopectin, pullulan, and related oligosaccharides, which enables a complete and efficient conversion of the branched polysaccharides into small fermentable sugars during saccharification process. The industrial manufacturing of glucose involves two successive enzymatic steps: liquefaction, carried out after gelatinisation by the action of α- amylase; saccharification, which results in further transformation of maltodextrins into glucose. -

OFFICIAL 27 October 2020 [139-20] Supporting document 1 Risk and Technical assessment – Application A1204 Βeta-amylase from soybean (Glycine max) as a processing aid (enzyme) Executive summary The purpose of the application is to amend Schedule 18 – Processing Aids of the Australia New Zealand Food Standards Code (the Code) to include the enzyme beta-amylase (β- amylase) (EC 3.2.1.2) produced from soybeans (Glycine max). β-Amylase is proposed as a processing aid in starch processing for the production of maltose syrup. The evidence presented to support the proposed use of the enzyme provides adequate assurance that the enzyme, in the quantity and form proposed to be used, is technologically justified and has been demonstrated to be effective in achieving its stated purpose. The enzyme meets international identity and purity specifications. β-Amylase from soybean is derived from the edible parts of the Glycine max plant, for which a history of safe use over generations is well known. FSANZ considers that soybean β-amylase is unlikely to pose an allergenicity concern. Bioinformatic analysis identified a degree of amino acid sequence homology between β- amylase from soybean and an allergenic protein from wheat, but FSANZ does not consider β-amylase to be of allergenic concern in wheat allergic individuals given the likely very low exposure and that the enzyme is likely to be digested in the stomach like other dietary proteins. The WHO/IUIS Allergen Nomenclature Database lists seven soy proteins that are food allergens. β-amylase from soybean is not one of these seven allergenic soy proteins and is not an allergen to individuals with soybean food allergy. -

In this study, the gene named IbSSI, was cloned from sweet potato, an important starch crop. A high expression level of IbSSI was detected in the leaves and storage roots of the sweet potato. Its overexpression significantly increased the content and granule size of starch and the proportion of amylopectin by up- regulating starch biosynthetic genes in the transgenic plants compared with wild-type plants (WT) and RNA interference plants. The frequency of chains with degree of polymerization (DP) 5–8 decreased in the amylopectin fraction of starch, whereas the proportion of chains with DP 9–25 increased in the IbSSI-overexpressing plants compared with WT plants. Further analysis demonstrated that IbSSI was responsible for the synthesis of chains with DP ranging from 9 to 17, which represents a different chain length spectrum in vivo from its counterparts in rice and wheat. These findings suggest that theIbSSI gene plays important roles in determining the content, composition, granule size and structure of starch in sweet potato. This gene may be utilized to improve the content and quality of starch in sweet potato and other plants. In plants, starch consists of amylose and amylopectin. Amylose mainly comprises linear chains that are linked by α-1, 4 O-glycosidic bonds, whereas amylopectin is highly branched and contains 5–6% α-1,6 O-glycosidic bonds to generate glucan branches of various lengths1. -
