Spirochaeta thermophila Assay conditions Catalogue number(s): CZ00921, 1 mg To recover maximal GFP-CBM64 activity, centrifuge a CZ00922, 3 × 1 mg required volume of the precipitated protein suspension (13000 xg for 5 min), remove the ammonium sulphate supernatant and ressuspend the resulting pellet in the same volume of 20 mM Tris-HCl, pH 7.5, 20 mM NaCl, 5 mM CaCl2. Description Proceed with the assay as required. GFP-CBM64 is a Carbohydrate Binding Protein derived from Spirochaeta thermophila. Recombinant GFP-CBM64, purified from Escherichia coli, is a modular protein containing a family 64 Carbohydrate Binding Module (CBM64) and an N-terminal Green Fluorescent Protein (GFP) (www.cazy.org). This GFP protein derivative is particularly recommended for subcellular localization studies. The protein is provided in 3.2 M ammonium sulphate at a 1 mg/mL concentration. Bulk Quality control assay quantities of this product are available on request. Protein purity is >90% as judged by SDS-PAGE followed by BlueSafe staining (MB15201). Electrophoretic Purity GFP-CBM64 purity was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) followed by BlueSafe staining (MB15201) (Figure 1).
