More specifically, it stimulates secretion of glucocorticoids such as cortisol, and has little control over secretion of aldosterone, the other major steroid hormone from the adrenal cortex. ACTH is secreted from the anterior pituitary in response to corticotropin-releasing hormone from the hypothalamus. The corticotropin-releasing hormone is secreted in response to many types of stress. Specificity and Preparation: This antibody recognizes mammalian ACTH. The immunogen was raised in rabbits against the synthetic human ACTH 1-39. The antibody is routinely tested by dot blot. Usage and Storage: Applications include immunostaining (1:200 to 1:600)1, immunoblotting (ATS in-house; 1:500), immunohistochemistry (fresh or frozen non-fixed sections; personal communication) and radioimmunoassay.2 Store the antibody at 4°C for one month or -20°C in undiluted aliquots for up to one year. Avoid repeated freezing and thawing. Gently spin down material before use; 5-10 seconds in a microfuge should be adequate. References: 1.
