CLCA1 belongs to the calcium-activated chloride channel regulator (previously known as chloride channel calcium activated or CLCA) family of self-cleaving, secreted metalloproteases that activate calcium-dependent chloride currents (ICaCC) in mammalian cells. Originally thought to be calcium-activated chloride channels (CaCCs), CLCA proteins were later identified as secreted CaCCs modulators. TMEM16A (a.k.a. Anoctamin1 and DOG1) is a CaCC expressed in airway epithelia and smooth muscle. The secreted CLCA1 is reported to physically interact with TMEM16A and modulate TMEM16A- dependent calcium-activated chloride currents. Adding CLCA1 to cells causes more TMEM16A channels to appear in the cell membrane, which increases the flow of chloride ions.
