Biotinylated Mouse Fc gamma RI / CD64 Catalog # AMS.CD4-M52E8-200ug For Research Use Only

Description Source Biotinylated Mouse Fc gamma RI / CD64 Protein (CD4-M52E8) is expressed from human HEK293 cells. It contains AA Glu 25 - Pro 297 (Accession # NP_034316).

Predicted N-terminus Glu 25 Molecular This protein carries a 15 AA fragment at the C-terminus, followed by a polyhistidine tag. There is one single lysine residue Characterization within the 15 AA fragment that is enzymatically labeled with biotin in vivo. The protein has a calculated MW of 33 kDa. The predicted N-terminus is Glu 25. The reducing (R) protein migrates as 45-60 kDa in SDS-PAGE due to glycosylation.

Endotoxin Less than 1.0 EU per μg of the Biotinylated Mouse Fc gamma RI by the LAL method. Purity >92% as determined by SDS-PAGE. Bioactivity The bioactivity is measured by its binding ability to mouse IgG2a in a SPR assay. Immobilized Biotinylated Mouse Fc gamma RI / CD64 Protein can bind mouse IgG2a with affinity constant of around 6.0 nM. Measured by its binding ability in a functional ELISA.Immobilized Yervoy Ipilimumab (Human IgG1) at 10 μg/mL (100 μL/well) can bind Biotinylated Mouse Fc gamma RI (Cat# CD4-M52E8 ) with a linear range of 0.03-0.5 μg/mL.

Formulation and Storage Formulation Lyophilized from 0.22 μm filtered solution in PBS, pH7.4. Normally Mannitol or Trehalose are added as protectants before lyophilization. Contact us for customized product form or formulation. Reconstitution See Certificate of Analysis for reconstitution instructions and specific concentrations. Storage Lyophilized Protein should be stored at -20℃ or lower for long term storage. Upon reconstitution, working aliquots should be stored at -20℃ or -70℃. Avoid repeated freeze-thaw cycles. No activity loss was observed after storage at: ● 4-8℃ for 12 months in lyophilized state; ● -70℃ for 3 months under sterile conditions after reconstitution. Background Background Receptors that recognize the Fc portion of IgG are divided into three groups designated Fc gamma RI, RII, and RIII, also known respectively as CD64, CD32, and CD16. Fc gamma RI binds IgG with high affinity and functions during early immune responses. Fc gamma RII and RIII are low affinity receptors that recognize IgG as aggregates surrounding multivalent antigens during late immune responses. High affinity immunoglobulin gamma Fc I is also known as FCGR1A, FCG1, FCGR1, CD64 and IGFR1, is a type of integral membrane that binds monomeric IgG-type with high affinity, which belongs to the immunoglobulin superfamily or FCGR1 family. FCGR1A / CD64 contains 3 Ig-like C2-type (immunoglobulin-like) domains. CD64 is constitutively found on only and monocytes, but treatment of polymorphonuclear leukocytes with cytokines like IFNγ and G-CSF can induce CD64 expression on these cells.
