Abstract: Saffron contained more than 150 volatile and several nonvolatile compounds, approximately 40–50 constituents have already been identified. It contained apocarotinoid glycosides: in particular crocin (crocetin- betadigentiobioside), colored intensive yellow orange; picrocrocin (glycosidic bitter principle, up to 4%): the apocarotinoids and picrocrocin were presumably breakdown products of a carotinoid-digentiobioside- diglucoside (protocrocin); volatile oil (0.4 to 1.3%): [(4,5-dehydro-betacyclocitral (safranal), 4-hydroxy-beta- cyclocitral (breakdown products of the picrocrocin)]; carotinoids: lycopene, alpha-, beta-, gamma-carotene; fatty oil and starch. The previous pharmacological studies revealed that saffron possessed antidepressant, anticonvulsant, antianxiety, memory improvement, for the treatment of tremor and morphine-withdrawal syndrome, antidiabetic, antioxidant, dermatological, immunological, cardiovascular, respiratory, reproductive, gastrointestinal, smooth muscle relaxation, anticancer, antiparasitic, anti-inflammatory, analgesic, protective ( hepatic, renal, CNS) and many other pharmacological effects. (Iridaceae) is monophyletic and contains about 100 species throughout the world. Crocus species have horticultural, medicinal and pharmacological importance. Saffron is the dried styles of C. sa- tivus and is one of the world’s most expensive spices by weight. Controversy exits about the taxonomy of the ge- nus and the species relationship. Exploring genetic diversity and inter-specific cross-ability are important tasks for conservation of wild taxa and for breeding of cultivated C. sativus. The present study was performed to study ge- netic variability and population structure in five Crocus L. species including Crocus almehensis Brickell & Mathew, C. caspius Fischer & Meyer, C. speciosus Marschall von Biberstein, C. haussknechtii Boissier, and C. sativus L. by inter simple sequence repeat (ISSR) molecular markers. We also used published internal transcribed spacer (ITS) sequences to study species relationship and compare the results with ISSR data. The results revealed a high degree of genetic variability both within and among the studied species. Neighbor joining (NJ) tree and network analysis revealed that ISSR markers are useful in Crocus species delimitation. Population fragmentation occurred in C. caspius and C. sativus. Both ISSR and sequenced based analyses separated C. -

A total of 369 polymorphic AFLP bands were generated and scored as binary data. Genetic similarities were determined. Cluster analysis revealed 4 major groups among the 26 genotypes examined in this study. The nuclear DNA contents (2C) of the 26 Crocus specimens were found to range from 5.08 pg in C. asumaniae to 9.75 pg in C. sativus. Polymorphic information content (PIC) values were used to examine the capacity of the various primer pairs to amplify polymorphisms in the Crocus specimens. -
