Blanchard, DePauw University As is well known, tryptophane is an amino acid derived from certain proteins and one of the few amino acids indispensable in the nutrition of higher animals. It was first isolated by Hopkins and Cole (1) in 1901 and afterwards synthesized by Ellinger and Flammand (2) in 1907. Under bacterial action it yields tryptamine, and in the body it undergoes changes which give rise to many substances, some of the most important being illustrated in the following scheme: CH 2 C00H\, /7\ CH 2 CH 2 C00H The point of particular interest to us just now is the relation of tryptophane to several vegetable alkaloids, and the recent discovery that the powerful alkaloid-like substances found in frogs and commonly known as toad poisons are also tryptophane derivatives. Indeed, it ap- pears that our generally accepted classification of vegetable alkaloids might well be slightly modified to provide for a division to be designated as Tryptophane Alkaloids. Owing to its widespread occurrence in nature, its intense pharmacological activity, and its presumably high biological significance, 5-hydroxytryptamine constitutes by far the most important member of this group of substances. It is not surprising, therefore, to find most of the space in the present paper devoted to it. This seems to be the first review on indolealkylamines. It calls attention to a number of experimental data and well established facts, but, more important, it shows the vast amount of work that remains to be done in this extremely fascinating field and the many problems which await solution. The current study aimed to evaluate its antinociceptive effects in bone cancer pain and explore the underlying mechanisms. Methods: Rat bone cancer model was used in this study. The withdrawal threshold evoked by stimulation of the hindpaw was determined using a 2290 CE electrical von Frey hair. The β-endorphin and IL-10 levels were measured in the spinal cord and cultured primary microglia, astrocytes, and neurons. Results: Cinobufagin, given intrathecally, dose-dependently attenuated mechanical allodynia in bone cancer pain rats, with the projected Emax of 90% MPE and ED50 of 6.4 μg. Intrathecal cinobufagin also stimulated the gene and protein expression of IL-10 and β-endorphin (but not dynorphin A) in the spinal cords of bone cancer pain rats. In addition, treatment with cinobufagin in cultured primary spinal microglia but not astrocytes or neurons stimulated the mRNA and protein expression of IL-10 and β-endorphin, which was prevented by the pretreatment with the IL- 10 antibody but not β-endorphin antiserum. Furthermore, spinal cinobufagin-induced mechanical antiallodynia was inhibited by the pretreatment with intrathecal injection of the microglial inhibitor minocycline, IL-10 antibody, β- endorphin antiserum and specific μ-opioid receptor antagonist CTAP.

Previous studies have clarified that psychoactive mushrooms produce psychoactive agents such as psilocybin, psilocin, ibotenic acid, and muscimol. However, the status of psychoactive mushrooms in most countries as illegal hallucinogens has prevented full investigation of their biochemical properties. Recent studies have shown that many psychoactive agents pass through the blood-brain barrier and act on neu- rotransmitter receptors. Psilocybin and psilocin are 5-HT1A and 5-HT2A/C receptor agonists, respectively, while ibotenic acid is a glutamic acid receptor agonist and muscimol is a GABAA receptor agonist. A new psychoac- tive agent, aeruginascin, has also been isolated from psychoactive mushrooms, and it is expected that more useful compounds will be discovered as the technology of component analysis advances. In addition, it has been shown that psilocybin and psilocin have high therapeutic efficiency for obsessive-compulsive disorder, which is a difficult-to-treat nervous disease. The increase of nervous diseases in modern society has thus given new importance to psychoactive mushrooms. In this review, we summarize the history of the use of psychoactive mushrooms, from pre-history to the modern age, describe their classification and distribution, survey previous studies, and discuss their therapeutic effects for difficult-to-treat nervous disease.

Recent studies have shown more than 100 such chemical components, including peptides, steroids, indole alkaloids, bufogargarizanines, organic acids, and others, in the parotoid and skins gland secretions from different species of toads. In traditional Chinese medicine (TCM), processed toad toxins have been used for treating various diseases for hundreds of years. Modern studies, including both experimental and clinical trials, have also revealed the molecular mechanisms that support the development of these components into medicines for the treatment of inflammatory diseases and cancers. More recently, there have been studies that demonstrated the therapeutic potential of toxins from other species of toads, such as Australian cane toads. Previous reviews mostly focused on the pharmaceutical effects of the whole extracts from parotoid glands or skins of toads. However, to fully understand the molecular basis of toad toxins in their use for therapy, a comprehensive understanding of the individual compound contained in toad toxins is necessary; thus, this paper seeks to review the recent studies of some typical compounds frequently identified in toad secretions.
