Structure of cell membrane ➢The structure of bacterial plasma membrane is that of unit membrane, i.e., a fluid phospholipid bilayer, composed of phospholipids (40%) and peripheral and integral proteins (60%) molecules. ➢The phospholipids of bacterial cell membranes do not contain sterols as in eukaryotes, but instead consist of saturated or monounsaturated fatty acids (rarely, polyunsaturated fatty acids). ➢Many bacteria contain sterol-like molecules called hopanoids. ➢The hopanoids most likely stabilize the bacterial cytoplasmic membrane. ➢The phospholipids are amphoteric molecules with a polar hydrophilic glycerol "head" attached via an ester bond to two non-polar hydrophobic fatty acid tails. ➢The phospholipid bilayer is arranged such that the polar ends of the molecules form the outermost and innermost surface of the membrane while the non-polar ends form the center of the membrane Fluid mosaic model ➢The plasma membrane contains proteins, sugars, and other lipids in addition to the phospholipids. ➢The model that describes the arrangement of these substances in lipid bilayer is called the fluid mosaic model ➢Dispersed within the bilayer are various structural and enzymatic proteins, which carry out most membrane functions. ➢Some membrane proteins are located and function on one side or another of the membrane (peripheral proteins). -

It is connected to the membrane-anchored rotary bacterial flagellar motor through a short hook. The bacterial flagellar filament consists of approximately 20,000 flagellins and can be several micrometers long. In this article, we reviewed the experimental works and models of flagellar filament construction and the recent findings of flagellar filament ejection during the cell cycle. The length-dependent decay of flagellar filament growth data supports the injection-diffusion model. The decay of flagellar growth rate is due to reduced transportation of long-distance diffusion and jamming. However, the filament is not a permeant structure. Several bacterial species actively abandon their flagella under starvation. Flagellum is disassembled when the rod is broken, resulting in an ejection of the filament with a partial rod and hook. The inner membrane component is then diffused on the membrane before further breakdown. These new findings open a new field of bacterial macro-molecule assembly, disassembly, and signal transduction. The coordinated assembly of flagellum and FAZ, during the cell cycle and the life cycle development, plays a pivotal role in organelle positioning, cell division and cell morphogenesis. To understand how the flagellum and FAZ assembly are coordinated, we examined the domain organization of the flagellum adhesion protein 1 (FLA1), a glycosylated, transmembrane protein essential for flagellum attachment and cell division. By immunoprecipitation of a FLA1-truncation mutant that mislocalized to the flagellum, a novel FLA1-binding protein (FLA1BP) was identified in procyclic Trypanosoma brucei. The interaction between FLA1 on the cell membrane and FLA1BP on the flagellum membrane acts like a molecular zipper, joining flagellum membrane to cell membrane and linking flagellum biogenesis to FAZ elongation. By coordinating flagellum and FAZ assembly during the cell cycle, morphology information is transmitted from the flagellum to the cell body. Review Questions Plasma Membrane 1. What is the function of the plasma membrane? The plasma membrane forms the outer boundary of all cells. Described as semi-permeable, the membrane regulates the passage of atoms and molecules in and out of the cell. All membrane-bound organelles are also built of plasma membrane. 2. Draw a phospholipid bilayer and label the polar heads and the non-polar tails. The plasma membrane is made of a phospholipid bilayer. Phospholipids have a glycerol backbone attached to two fatty acid chains (one is unsaturated) and one phosphate group. The phosphate has a negative charge. The rest of the lipid is nonpolar. Water is attracted to the phosphate group and repelled by the rest of the molecule, so a phospholipid has a dual nature: a hydrophilic region (“head”) and a hydrophobic region (“tails”). When placed in water, the phospholipids form a bilayer. The heads face outward and the tails stay inside. The bilayer is semi-permeable barrier. Some substances can freely cross the membrane whereas others are stopped. 3. Why is the plasma membrane described as a “fluid mosaic”? The plasma membrane is a fluid because the phospholipid molecule is unsaturated. At room temperature, this causes the bilayer to have the consistency of salad oil. The term “mosaic” refers to the variety of embedded transmembrane proteins scattered throughout the membrane. 4. Why is cholesterol an important component of animal cell membranes? Cholesterol is a component of the cell membranes of animals. Cholesterol makes the membrane less fluid and therefore more impermeable to biological molecules. In a sense, cholesterol creates a more solid membrane and a more restrictive membrane. -

Explain the main functions of the cell membrane and the cell wall. Describe what happens during diffusion and osmosis. Describe what happens during active and passive transport. All cells are surrounded by a thin, flexible barrier known as the cell membrane. Many cells also produce a strong supporting layer around the membrane known as the cell wall. The cell membrane regulates what enters and leaves the cell and also provides protection and support (maintains homeostasis). Nearly all cell membranes are made of a double- layered sheet called a phospolipid bilayer. A phospholipid is made of a polar head which is hydrophilic or “water loving” and two nonpolar fatty acid chains or tails that are hydrophobic or “water fearing”. The phospholipids organize themselves in a bilayer to hide their hydrophobic tails from and expose their hydrophilic heads to the water found on both sides of the cell membrane. In addition to the lipids, most cell membranes contain protein molecules that are embedded in the lipid bilayer. Some of these proteins form channels and pumps that help move material across the cell membrane. Cell walls are present in plants, algae, fungi, and many prokaryotes (but not animals). Cell walls lie outside the cell membrane. Most cell walls are porous enough to allow water, oxygen, carbon dioxide, and certain other substances through easily. The main function of the cell wall is to provide support and protection for the cell. Most cell walls are made from fibers of carbohydrates and protein. These substances are produced inside the cell and released at the surface of the cell membrane where they are assembled to form the wall.
