Parasites Vectors (2019) 12:553 https://doi.org/10.1186/s13071-019-3809-9 Parasites & Vectors RESEARCH Open Access Molecular characterization and genetic diversity of Ornithonyssus sylviarum in chickens (Gallus gallus) from Hainan Island, China Biswajit Bhowmick1, Jianguo Zhao1, Øivind Øines2, Tianlin Bi1, Chenghong Liao1, Lei Zhang1 and Qian Han1* Abstract Background: The northern fowl mite (NFM), Ornithonyssus sylviarum, is an obligatory hematophagous ectoparasite of birds and one of the most important pests in the poultry industry on several continents. Although NFM poses a serious problem, it remains a neglected pest of poultry in China and other Asian countries. Therefore, a molecular analysis was conducted to provide baseline information on the occurrence, genetic diversity and emergence of NFM in poultry farms from China. Methods: This study focused on morphological description and identifcation of adults based on electron micros- copy, molecular sequencing of the mitochondrial cox1 gene and phylogenetic analysis. We have also used the DNA sequences of the cox1 gene to study the genetic diversity, population structure and demographic history. The neutral- ity tests were used to analyze signatures of historical demographic events. Results: The mites collected were identifed as the northern fowl mite Ornithonyssus sylviarum based on external morphological characterization using electron microscopy. Molecular analysis using a 756-bp long partial fragment of the cox1 gene revealed 99–100% sequence identity with NFM and phylogenetic inferences showed a bootstrap value of 99% indicating a well-supported monophyletic relationship. Molecular diversity indices showed high levels of hap- lotype diversity dominated by private haplotypes, but low nucleotide divergence between haplotypes. The Tajima’s D test and Fu’s Fs test showed negative value, indicating deviations from neutrality and both suggested recent popu- lation expansion of mite populations supported by a star-like topology of the isolates in the network analysis. Our genetic data are consistent with a single introduction of NFM infestations and the spread of NFM infestation in Hainan poultry farms and a private haplotype dominance, which suggest that infestations are recycled within the farms and transmission routes are limited between farms. Conclusions: To our knowledge, this is the frst time a molecular report of NFM in chicken from China including other Asian countries using DNA barcoding. The fndings have potential implications with respect to understanding the transmission patterns, emergence and populations trends of parasitic infestations of poultry farms that will help for setting the parameters for integrated pest management (IPM) tactics against mite infestations. However, this mite can affect several vertebrate hosts, including humans, leading to dermatitis, pruritus, allergic reactions, and papular skin lesions. This study evaluated the epidemiological characteristics of O. sylviarum attacks on poultry workers, including data on laying hens, infrastructure and management of hen houses, and reports of attacks by hematophagous mites. In addition, a case of mite attack on a farm worker on a laying farm in the Midwest region in Minas Gerais is presented. It was found that 60.7% farm workers reported attacks by hematophagous mites. Correspondence analysis showed an association between reports of mite attacks in humans with (1) presence of O. sylviarum in the hen house, (2) manual removal of manure by employees, and (3) history of acaricide use.

Cromroy2 Introduction The tropical fowl mite, commonly found on birds, has become a pest to people in areas of high bird populations or where birds are allowed to roost on roofs, around the eaves of homes, and office buildings. Nesting birds are the worst offenders. After the birds abandon their nests, the mites move into the building through windows, doors, and vents and bite the occupants. The bite is irritating, and some individuals react to the bite with prolonged itching and painful dermatitis. Several to many reports are received each year of mites invading homes. The mites are usually the tropical fowl mite found in the central and southern areas of the state. O objetivo deste estudo foi relatar as espécies de ácaros hematófagos parasitos de Passer domesticus (Linnaeus, 1758) e analisar os índices de infestação em relação ao gênero, massa corporal e comprimento total dos hospedeiros. Para isso, cada um dos 100 pardais capturados na área urbana de Pelotas, Rio Grande do Sul, Brasil, foi sexado, pesado e medido. A coleta dos ácaros foi realizada após aplicação de um talco repelente de ectoparasitos sobre o corpo das aves. Pellonyssus reedi ocorreu em 29 pardais com intensidade média de 8,37 ácaros/hospedeiro e Ornithonyssus bursa ocorreu em duas aves, somente um hospedeiro macho adulto apresentou co-infestação. A prevalência e intensidade média de infestação por P. reedi entre hospedeiros machos e fêmeas adultos não apresentou diferença significativa, também não houve correlação entre a abundância dessa espécie, massa corporal e comprimento total dos pardais.

Entomol. 44(4): 709Ð713 (2007) ABSTRACT The northern fowl mite, Ornithonyssus sylviarum (Canestrini and Fanzago, 1877) (Mesostigmata: Macronyssidae) is a broadly distributed blood-feeding parasite that has been collected from many birds of temperate regions. Previously, the most complete host list was published in 1938, and it included 15 North American (Canada, United States, and Mexico) host species. In the process of a general survey of bird-associated mites in Alberta, Canada, we recovered many O. sylviarum specimens. Herein, we update the previous host list with these observations and records published since 1938. We collected mites by washing the bodies of salvaged birds and examining the Þltrate. Northern fowl mites were collected from 26 host species, with 16 of these species being the Þrst host records for North America. Including results from the current study, O. sylviarum has been reported from 72 species of North American birds from 26 families. This updated host list will be useful to anyone interested in the role of O. sylviarum in transmission of avian disease. Methods: Clinically healthy, naturally mite-infested laying hens (n = 132), approximately 32 weeks of age, were ranked by Day -9 mite vent counts and randomized among 12 study pens, each to hold one of four treatment groups. Three groups received fluralaner-medicated water by oral gavage at dose rates of 0.25, 0.5 or 1.0 mg/kg on Days 0 and 7; one group was an untreated control (three pens for each group). Five naturally infested untreated birds were included in each pen to act as mite-infested source birds. Thus each pen, treated and control, had six non-source birds for assessment of efficacy, plus five source birds to provide ongoing challenge. Primary efficacy assessments were based on mean O. sylviarum vent counts from non-source birds in the control and treated group pens on Days 1, 2, 6, 8, 12, 15, 19, 22 and 26. A case of infestation with trop- 33 ical rat mites in a family is reported here. Three mice that had been removed 34 from the home two months before were the reservoir. The mites were detect- 35 ed in a room where the cage with the mice had been placed months ago. 36 Treatment requires the eradication of the parasites on its hosts (by a veterinar- 37 ian) and in the environment (by an exterminator) with adequate acaricides 38 such as permethrin.
