This study was focused on H biotechnological production tools of R. stricta, namely undifferentiated cell cultures, and an Agrobacterium rhizogenes- mediated transformation method to obtain hairy roots expressing heterologous genes from the early TIA pathway. Rha zya alkaloids comprise a wide range of structures and polarities, therefore, many A analytical methods were developed to investigate the alkaloid l k contents in in vitro cultures. Targeted and non-targeted analyses a l o were carried out using gas chromatography-mass spectrometry i d (GC-MS), high performance liquid chromatography (HPLC), ultra s o performance liquid chromatography-mass spectrometry (UPLC- f i MS) and nuclear magnetic resonance (NMR) spectroscopy. n Calli derived from stems contained elevated levels of v i t r strictosidine lactam compared to other in vitro cultures. It o was revealed that only leaves were susceptible to Agrobacterium c u infection and subsequent root induction. The transformation l t u efficiency varied from 22% to 83% depending on the gene. A total r e of 17 TIAs were identified from hairy root extracts by UPLC-MS. -

Summary: Callus was produced from hypocotyl sections of in vitro germinated seedlings of Cynanchum acutum on Murashige and Skoog media (1962) containing 0.5 - 4.0 mg/l a-Naphthalene acetic acid in combination with 0.2-1.0 mg/l Benzyl adenine. After 12 weeks in culture both fresh and dry weights (g) were significantly higher on 0.5 mg/l NAA and 0.2 mg/l BA. Cardiac glycoside concentration was highest (1.2 mg/g DW) on the same growth regulators combination. Flavonoids were highest (0.198 mg/g) on 0.5mg/l NAA and 1.0 mg/l BA. Twenty eight weeks old callus gave highest cardiac glycosides (1.3 mg/g) on media lacking BA and 0.5 mg/l NAA. Flavonoids concentration (0.27 mg/g) was highest on the same auxin concentration in the presence of 0.2 mg/l BA. Regeneration from callus cultures was obtained when 8-weeks old callus was subcultured on MS hormone free media for 6 weeks. Shoots were rooted on MS supplemented with 0.1 mg/l NAA and acclimatized in growth chamber. Adventitious shoots showed significantly higher CG (6.5 and 4.3 mg/g) than the wild plants (2.8), while regenerants gave comparable concentration (2.3) to the wild. -

In order to identify processes attributable to its adaptation to this harsh environment, we profiled the foliar transcriptome of apical and mature leaves harvested from the field at three time periods of the same day. Results: Next generation sequencing was used to reconstruct the transcriptome and quantify gene expression. 28018 full length transcript sequences were recovered and 45.4% were differentially expressed (DE) throughout the day. We compared our dataset with microarray experiments in Arabidopsis thaliana (Arabidopsis) and other desert species to identify trends in circadian and stress response profiles between species. 34% of the DE genes were homologous to Arabidopsis circadian-regulated genes. Independent of circadian control, significant overlaps with Arabidopsis genes were observed only with heat and salinity/high light stress-responsive genes. Also, groups of DE genes common to other desert plants species were identified. We identified protein families specific to R. stricta which were found to have diverged from their homologs in other species and which were over -expressed at midday. Conclusions: This study shows that temporal profiling is essential to assess the significance of genes apparently responsive to abiotic stress. -

In this genus, 16 species are of high medicinal value. The extracts of the root and/or rhizome parts have been applied in traditional Chinese medicines (TCM) for the prevention and treatment of various illnesses for centuries. C21 steroids, as the typical constituents of Cynanchum species, possess a variety of structures and pharmacological activities. This review summarizes the comprehensive information on phytochemistry and pharmacology of C21 steroid constituents from Cynanchum plants, based on reports published between 2007 and 2015. Our aim is to provide a rationale for their therapeutic application, and to discuss the future trends in research and development of these compounds. A total of 172 newly identified compounds are reviewed according to their structural classifications. Their in vitro and in vivo pharmacological studies are also reviewed and discussed, focusing on antitumor, antidepressant, antifungal, antitaging, Na+/K+-ATPase inhibitory, appetite suppressing and antiviral activities. Future research efforts should concentrate on in vitro and in vivo biological studies and structure activity relationship of various C21 steroid constituents. It is compared with two morphologically similar species, Cynanchum decipiens and C. longipedunculatum. Cynanchum brevipedunculatum differs from the preceeding species in having much bigger leaves, adaxially sparsely strigillose, abaxially glabrous or sparsely strigillose on veins, shorter peduncle and the very distinctive bowl-shaped corona. A comprehensive morphological description of C. brevipedunculatum is provided, together with photographs, and a conservation assessment for this rare vine species. Pollen Transfer Efficiency of Apocynum Cannabinum (Apocynaceae): a Comparative Perspective

Abstract—Pollen transfer efficiency (PTE), the percentage of removed pollen delivered to conspecific stigmas, has been implicated in the morphological evolution, population dynamics, and lineage diversification of flowering plants. Pollinia, the aggregated contents of pollen sacs, present in Apocynaceae subfamilies Asclepiadoideae (milkweeds), Secamonoideae, and Periplocoideae and orchids (Orchidaceae), are the pre-eminent example of a plant trait that elevates PTE (to ca. 25%). However, comparison of species with pollinia to “average” flowers (PTE ca. 1%) may over-estimate the gains from pollinia. We hypothesize that elevated PTE evolved in Apocynaceae prior to pollinia. We measured PTE and pollen to ovule ratio, a possible correlate of PTE, in Apocynum cannabinum, a milkweed relative with pollen tetrads (instead of pollinia) and simple bands of style head adhesive (instead of complex pollinium-carrying translators), comparing them to reports of other species collated from the literature. PTE of A. cannabinum is 7.9%, in the 24th percentile of reports for 36 milkweed species, but more than twice the highest PTE reported for a species with monads (3.4%). The bands of style head adhesive are functionally equivalent to the translators of milkweeds. The pollen to ovule ratio of A. cannabinum, at 19.8, is in the 94th percentile of ratios reported for milkweeds (mean 9.6). -
