Long-term plasticity of interneurons has mostly been studied in unidentified GABAergic cells and is known to be very heterogeneous. Here we tested whether cell type-specific plasticity properties in distinct GABAergic interneuron types might underlie this heterogeneity. We show that long-term potentiation (LTP) and depression (LTD), two common forms of synaptic plasticity, are expressed in a highly cell type-specific manner at glutamatergic synapses onto hippocampal GABAergic neurons. Both LTP and LTD are generated in interneurons expressing parvalbumin (PVϩ), whereas interneurons with similar axon distributions but expressing cannabinoid receptor-1 show no lasting plasticity in response to the same protocol. In addition, LTP or LTD occurs in PVϩ interneurons with different efferent target domains. Perisomatic-targeting PVϩ basket and axo-axonic interneurons express LTP, whereas glutamatergic synapses onto PVϩ bistratified cells display LTD. Both LTP and LTD are pathway specific, independent of NMDA receptors, and occur at synapses with calcium-permeable (CP) AMPA receptors. Plasticity in interneurons with CP-AMPA receptors strongly modulates disynaptic GABAergic transmission onto CA1 pyramidal cells.

We examined the mRNA expression in brain regions to investigate how selective vulnerability could help explain the phenotypes and analysed the effect of mutations on patient-derived mRNA. Despite the discovery of more than 90 causative CMT genes through the advent and use of next-generation sequencing (NGS) technologies, there are still patients with specific subtypes of CMT, such as CMT2 and distal Hereditary Neuropathy (dHMN) that remain without a genetic diagnosis. Where contributions of others are involved, this has been clearly indicated in the text. 2 Abstract Channelopathies are disorders caused by inherited mutations of specific ion channels. Neurological channelopathies in particular offer a window into fundamental physiological functions such as action potential modulation, synaptic function and neurotransmitter release. One such channelopathy Episodic Ataxia type 1 (EA1), is caused by a mutation to the gene that encodes for the potassium channel subunit Kv1.1. This channel is predominantly found in presynaptic terminals and EA1 mutations have previously been shown to result in increased neuronal excitability and neurotransmitter release. A possible reason is that presynaptic action potential waveforms are affected in EA1. Thus far, direct electrophysiological recording of presynaptic terminals has been limited to large specialised synapses e.g. mossy fibre boutons, or axonal blebs, unnatural endings of transected axons. This is not representative of the vast majority of small synapses found in the forebrain. Using a novel technique termed Hopping Probe Ion Conductance Microscopy (HPICM) I have been able to directly record action potentials from micrometer sized boutons in hippocampal neuronal culture. I have shown that in a knockout model of Kv1.1 and in a knockin model of the V408A EA1 mutation, presynaptic action potentials are broader than in wild type; however action potentials are unaffected in the cell body.

TO: The Council of the Society for Neuroscience (SfN) FROM: Members of SfN (current and recently active) As members of SfN, we are proud of our society’s legacy of scientific excellence and leadership that was vividly on display at the recent 50th anniversary meeting. At the same time, this anniversary prompts us to consider how the Society can take a leading role in addressing the climate crisis which is a defining challenge for our generation. Climate scientists report that the earth has experienced approximately 1 degree C of anthropogenic global heating since pre-industrial times and that we are on track to reach 1.5 degrees C as soon as 20301. There is a consensus amongst climate scientists, ratified by governments, that we must substantially reduce the emissions of greenhouse gases in the next ~11 years to prevent increasingly dire conseQuences2. It is clear that unabated emissions threaten to increasingly disrupt our world, including our research science3. We accept the scientific consensus that there must now be a rapid decarbonization in all parts of society, including professional bodies of scientists such as SfN. The Society’s success and growth reQuires us to address the climate impact of ~30,000 scientists congregating for an annual meeting. We recognize that our conferences play a crucial and important role in neuroscience, providing a uniQue venue for interdisciplinary discussion and dissemination of the latest findings. The benefits of conferences include creation of long-term collaborations and friendships, career-building and raising the profile of science worldwide.

A seizure is a period of runaway electrical activity during which the normal balance between excitation and inhibition is lost. Current anti-seizure drugs either dampen excitatory mechanisms or boost inhibitory ones. Current gene-therapy strategies, by contrast, use harmless viruses to introduce one or two therapeutic genes into the defined volume of tissue from which focal epilepsy emanates. “It is more personal, more targeted, and prob- ably has fewer side effects because we treat the tissue that needs to be treated, instead of treating the whole body,” says Merab Kokaia, a neuroscientist working on this approach at Lund University in Sweden. RESTORING BALANCE The brains of people with epilepsy contain NEUROLOGY increased amounts of neuropeptide Y (NPY), a chemical that certain neurons release when they are especially active. During this period, we have initiated 15 new research projects, adding to 30 active projects spanning the clinical, educational and social elements of paediatric epilepsy. We have published 62 peer-reviewed items of primary research, 28 reviews and a further 5 chapters in books. We continue to share expertise with research and clinical colleagues and in January 2020 held our 10th International Paediatric Epilepsy Research Retreat for researchers and collaborators across the unit. This meeting was a wonderful celebration of the work of the unit over the past decade. We enjoyed record attendance as well as welcoming two eminent Research Moderators; Professor Alexis Arzimanoglou of University Hospitals of Lyon, and Professor Mathias Koepp of even greater still, the changes to all lives and pursuits UCL Queen Square Institute of Neurology.
