Recombinant Mouse and Activation Regulated Chem okine (CCL17)

Instruction Manual

Catalog Number D-68010 Description TARC, a CC , is predominantly produced by dendritic cells in the thymus and at a lower level in the lung, colon, and small intestine. TARC is in addition transiently expressed in stimulated peripheral blood mononuclear cells. Recombinant TARC has been shown to be chemotactic for T cell lines but not or neutrophils. CCL-17 was recently identified to be a specific functional ligand for CCR4, a receptor that is selectively expressed on T cells. CCL17 is one of quite a few Cys-Cys (CC) clustered on the q arm of 16. CCL17 shows chemotactic activity for T lymphocytes, but not monocytes or . CCL17 binds to chemokine receptors CCR4 and CCR8. This chemokine plays important roles in T cell development in thymus as well as in trafficking and activation of mature T cells. Recombinant mouse TARC produced in E. coli is an 7.9 kDa protein containing 70 amino acid residues. It has been purified by proprietary chromatographic techniques. Quantity 20 µg Molecular Mass 7.9 kDa Specific Activity see below Source E. coli Purity Greater than 97% (determined by SDS-PAGE and HPLC analysis). Endotoxin Level < 0.1 ng per µg (1EU/µg) of rMu TARC.

Biological Activity Fully biologically active when compared to a standard. The ED ₅₀ as determined by its ability to chemoattract BaF3 mouse pro-B cells transfected with human CCR4 is typically 2-10 ng/ml. Formulation White (freeze-dried) powder. Lyophilized from a sterile-filtered (0.2 µm) concentrated protein solution in 1×PBS (pH 7.4). Reconstitution Please Note: Always centrifuge product briefly before opening the vial. Reconstitute in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can be diluted into other aqueous buffers and stored at -20°C for future use. Storage & Stability The lyophilized powder, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted TARC should be used immediately or stored in undiluted working aliquots at -20°C. Avoid repeated freeze / thaw cycles.

FOR IN VITRO RESEARCH USE ONLY. NOT FOR DIAGNOSTIC OR THERAPEUTIC PROCEDURES.
