Foliar samples were collected in okra production fields and tested by using begomovirus universal primers. Begomovirus infection was confirmed in only one (#5157) out of 196 samples. Total DNA was subjected to PCR amplification and introduced into okra seedlings by a biolistic method; the bombarded DNA sample was infectious to okra plants. The DNA-A and DNA-B of isolate #5157 were cloned and their nucleotide sequences exhibited typical characteristics of New World bipartite begomoviruses. The DNA-A sequence shared 95.6% nucleotide identity with an isolate of Sida micrantha mosaic virus from Brazil and thus identified as its okra strain. The clones derived from #5157 were infectious to okra, Sida santaremnensis and to a group of Solanaceae plants when inoculated by biolistics after circularization of the isolated insert, followed by rolling circle amplification. Virus transcript abundance was assessed by quantifying selected gene transcripts of Tomato mottle virus (ToMoV, a New World bipartite begomovirus) and Tomato yellow leaf curl virus (TYLCV, an Old World monopartite begomovirus) in whiteflies (Bemisia tabaci biotype B) after feeding on virus-infected tomato plants and after subsequent transfer to cotton, a plant that is immune to the selected begomoviruses. Real-time RT-PCR was performed using specific primers for three ToMoV genes (AV1, BC1 and BV1) and three TYLCV genes (V1, V2 and C3). The ToMoV gene transcripts rapidly became undetectable in whiteflies following transfer from tomato to cotton, probably because degradation was not accompanied by new synthesis. On the other hand, TYLCV transcripts increased after transfer of whiteflies to cotton, indicating active TYLCV transcription. College, Dibrugarh - 786 001 (Assam), India. E-mail: [email protected] Abstract Changes in botanical names of flowering plants are an issue which comes up from time to time. While there are valid scientific reasons for such changes, it also creates some difficulties to the floristic workers in the preparation of a new flora. Further, all the important monumental floras of the world have most of the plants included in their old names, which are now regarded as synonyms. In north east India, “Flora of Assam” is an important flora as it includes result of pioneering floristic work on Angiosperms & Gymnosperms in the region. But, in the study of this flora, the same problems of name changes appear before the new researchers. Therefore, an attempt is made here to prepare an updated account of the new names against their old counterpts of the plants included in the first two volumes of the flora, on the basis of recent standard taxonomic literatures. In this, the unresolved & controversial names are not touched & only the confirmed ones are taken into account. In the process new names of 470 (four hundred & seventy) dicotyledonous plant species included in the concerned flora are found out. Most fermenters are operated at a mesophilic temperature regime. Modern trends in waste management aim to minimize waste generation. One of the strategies can be achieved by anaerobically stabilizing the sludge by raising the temperature. Higher temperatures will allow faster decomposition of organic matter, shortening the retention time, and increasing biogas production. This work is focused on the description of changes in the community of methanogenic microorganisms at different temperatures during the sludge stabilization. At higher temperatures, biogas contained a higher percentage of methane, however, there was an undesirable accumulation of ammonia in the fermenter. Representatives of the hydrogenotrophic genus Methanoliea were described at all temperatures tested. At temperatures up to 50 ◦C, a significant proportion of methanogens were also formed by acetoclastic representatives of Methanosaeta sp. and acetoclastic representatives of the order Methanosarcinales. -

www.nature.com/scientificreports OPEN Pan‑genome analysis and ancestral state reconstruction of class halobacteria: probability of a new super‑order Sonam Gaba1,2, Abha Kumari2, Marnix Medema 3 & Rajeev Kaushik1* Halobacteria, a class of Euryarchaeota are extremely halophilic archaea that can adapt to a wide range of salt concentration generally from 10% NaCl to saturated salt concentration of 32% NaCl. It consists of the orders: Halobacteriales, Haloferaciales and Natriabales. Pan‑genome analysis of class Halobacteria was done to explore the core (300) and variable components (Softcore: 998, Cloud:36531, Shell:11784). The core component revealed genes of replication, transcription, translation and repair, whereas the variable component had a major portion of environmental information processing. The pan‑gene matrix was mapped onto the core‑gene tree to fnd the ancestral (44.8%) and derived genes (55.1%) of the Last Common Ancestor of Halobacteria. A High percentage of derived genes along with presence of transformation and conjugation genes indicate the occurrence of horizontal gene transfer during the evolution of Halobacteria. A Core and pan‑gene tree were also constructed to infer a phylogeny which implicated on the new super‑order comprising of Natrialbales and Halobacteriales. Halobacteria1,2 is a class of phylum Euryarchaeota3 consisting of extremely halophilic archaea found till date and contains three orders namely Halobacteriales4,5 Haloferacales5 and Natrialbales5. Tese microorganisms are able to dwell at wide range of salt concentration generally from 10% NaCl to saturated salt concentration of 32% NaCl6. Halobacteria, as the name suggests were once considered a part of a domain "Bacteria" but with the discovery of the third domain "Archaea" by Carl Woese et al.7, it became part of Archaea. -

People all over the world are dying from multidrug resistant infections, and it is predicted that bacterial infections will once again become the leading cause of death. One human opportunistic pathogen of great concern is Pseudomonas aeruginosa. P. aeruginosa is the most abundant pathogen in cystic fibrosis (CF) patients’ lungs over time and is resistant to most currently used antibiotics. Chronic infection of the CF lung is the main cause of morbidity and mortality in CF patients. With the rise of multidrug resistant bacteria and lack of novel antibiotics, treatment for CF patients will become more problematic. Escalating the problem is a lack of research from pharmaceutical companies due to low profitability, resulting in a large void in the discovery and development of antibiotics. Thus, research labs within academia have played an important role in the discovery of novel compounds. Environmental bacteria are known to naturally produce secondary metabolites, some of which outcompete surrounding bacteria for resources. We hypothesized that environmental Pseudomonas from diverse soil and water habitats produce secondary metabolites capable of inhibiting the growth of CF derived P. aeruginosa. To address this hypothesis, we used a population based study in tandem with transposon mutagenesis and bioinformatics to identify eight biosynthetic gene clusters (BGCs) from four different environmental Pseudomonas strains, S4G9, LE6C9, LE5C2 and S3E10. -

Accepted: February 16, 2019 Abstract The last two steps of the purine biosynthetic pathway may be catalyzed by different enzymes in prokaryotes. The genes that encode these enzymes include homologs of purH, purP, purO and those encoding the AICARFT and IMPCH domains of PurH, here named purV and purJ, respectively. In Bacteria, these reactions are mainly catalyzed by the domains AICARFT and IMPCH of PurH. In Archaea, these reactions may be carried out by PurH and also by PurP and PurO, both considered signatures of this domain and analogous to the AICARFT and IMPCH domains of PurH, respectively. These genes were searched for in 1,403 completely sequenced prokaryotic genomes publicly available. Our analyses revealed taxonomic patterns for the distribution of these genes and anticorrelations in their occurrence. The analyses of bacterial genomes revealed the existence of genes coding for PurV, PurJ, and PurO, which may no longer be considered signatures of the domain Archaea. Although highly divergent, the PurOs of Archaea and Bacteria show a high level of conservation in the amino acids of the active sites of the protein, allowing us to infer that these enzymes are analogs.
