Prisinzano Date approved: July 18, 2012 ii ABSTRACT Kappa opioid (KOP) receptors have been suggested as an alternative target to the mu opioid (MOP) receptor for the treatment of pain because KOP activation is associated with fewer negative side-effects (respiratory depression, constipation, tolerance, and dependence). The KOP receptor has also been implicated in several abuse-related effects in the central nervous system (CNS). KOP ligands have been investigated as pharmacotherapies for drug abuse; KOP agonists have been shown to modulate dopamine concentrations in the CNS as well as attenuate the self-administration of cocaine in a variety of species, and KOP antagonists have potential in the treatment of relapse. One drawback of current opioid ligand investigation is that many compounds are based on the morphine scaffold and thus have similar properties, both positive and negative, to the parent molecule. Thus there is increasing need to discover new chemical scaffolds with opioid receptor activity. -

THE EFFECT OF THE FLAVONOIDS QUERCETIN AND GENISTEIN ON THE ANTIOXIDANT ENZYMES Cu, Zn SUPEROXIDE DISMUTASE, GLUTATHIONE PEROXIDASE, AND GLUTATHIONE REDUCTASE IN MALE SPRAGUE-DAWLEY RATS by ANNETTE CAIRNS GOVERNO (Under the Direction of Joan G. Fischer) ABSTRACT Quercetin (QC) and genistein (GS) are phytochemicals found in fruits and vegetables. These compounds may exert protective effects by altering antioxidant enzyme activities. The objective of the study was to examine the effects of QC and GS supplementation on the activities of the antioxidant enzymes glutathione reductase (GR), glutathione peroxidase (GSHPx), and Cu, Zn superoxide dismutase (SOD) in liver, and SOD activity in red blood cells (RBC), as well as the Ferric Reducing Antioxidant Potential (FRAP). Male, weanling Sprague-Dawley rats (n=7-8 group) were fed quercetin at 0.3, 0.6 or 0.9g/100g of diet or genistein at 0.008, 0.012, or 0.02g/100g diet for 14d. GS supplementation significantly increased liver GSHPx activity compared to control (p<0.01). GS did not significantly alter activities of liver SOD and GR, or RBC SOD. QC did not significantly alter antioxidant enzyme activities in liver or RBC. Neither QC nor GS increased the antioxidant capacity of serum. In conclusion, low levels of GS significantly increased liver GSHPx activity, which may contribute to this isoflavone’s protective effects. The thesis contains as its main content, work that has not been previously submitted for a degree at any other university. i Abstract The complementary and alternative medicines (CAM) industry is worth over US$110 billion globally. Products are available to consumers with little medical advice; with many assuming that such products are ‘natural’ and therefore safe. However, with adulterated, contaminated and fraudulent products reported on overseas markets, consumers may be placing their health at risk. Previous studies into product content have reported undeclared plant materials, ingredient substitution, adulteration and contamination. However, no large-scale, independent audit of CAM has been undertaken to demonstrate these problems in Australia. This study aimed to investigate the content and quality of CAM products on the Australian market. 135 products were analysed using a combination of next-generation DNA sequencing and liquid chromatography-mass spectrometry. Nearly 50% of products tested had contamination issues, in terms of DNA, chemical composition or both. 5% of the samples contained undeclared pharmaceuticals. -

The Ohio State University, 2008 Dr. Maureen Geraghty, Advisor This pilot study was a retrospective chart review. The purposes of this study were to describe the prevalence herbal supplement use, to identify characteristics linked to increased herbal supplement use and, to identify adverse outcomes linked to herbal supplement use. Rate of use in the study sample of 2136 charts was 1.1% and identified 17 supplements. The most common supplements identified were teas. Characteristics of the neonates and controls were analyzed as appropriate and revealed no statistical significance. Characteristics of the mothers also revealed no statistical difference. There was a statistically significant difference between herbal users and herbal non-users and the trimester prenatal care began. Neonatal outcomes were statistically different on two measures. Food is considered as the main source of exposure to BPA as it leaches out from the food containers as well as surface coatings into it. BPA is toxic to vital organs such as liver kidney and brain. Quercetin, the most abundant flavonoid in nature, is present in large amounts in vegetables, fruits and tea. The aim of the present study was to evaluate the toxic effects of BPA in prostate gland and seminal vesicle of mice and its possible amelioration by quercetin. Material and methods: Inbred Swiss strain male albino mice were orally administered with BPA (80, 120 and 240 mg/kg body weight/day) for 45 Days. Oral administration of BPA caused significant, dose-dependent reduction in absolute and relative weights of prostate gland and seminal vesicle. Results and conclusion: Biochemical analysis revealed that protein content reduced significantly, whereas acid phosphatase activity increased significantly in prostate gland and reduction in fructose content was observed in seminal vesicle. Oral administration of quercetin (30, 60 and 90 mg/kg body weight/day) alone with high dose of BPA (240 mg/kg body weight/day) for 45 days caused significant and dose-dependent amelioration in all parameters as compared to BPA along treated group. -

Batch specific analytical results are provided on each certificate of analysis. Laboratory Procedures (−)-Epigallocatechin gallate (EGCG) is supplied as a crystalline solid. A stock solution may be made by dissolving the EGCG in an organic solvent purged with an inert gas. EGCG is soluble in organic solvents such as ethanol, DMSO, and dimethyl formamide. The solubility of EGCG in these solvents is approximately 20, 25, and 30 mg/ml, respectively. Further dilutions of the stock solution into aqueous buffers or isotonic saline should be made prior to performing biological experiments. Ensure that the residual amount of organic solvent is insignificant, since organic solvents may have physiological effects at low concentrations. Organic solvent-free aqueous solutions of EGCG can be prepared by directly dissolving the crystalline compound in aqueous buffers. The solubility of EGCG in PBS (pH 7.2) is approximately 25 mg/ml. In vivo studies showed that BPA exposure has deleterious effects on spermatogenesis by disturbing the hypothalamic–pituitary–gonadal axis and inducing oxidative stress in testis. This compound seems to disrupt hormone signalling even at low concentrations, modifying the levels of inhibin B, oestra- diol, and testosterone. The adverse effects on seminal parameters are mainly supported by studies based on urinary BPA concentration, showing a negative association between BPA levels and sperm concentration, motility, and sperm DNA damage. Recent studies explored potential approaches to treat or prevent BPA-induced testicular toxicity and male infertility. Since the effect of BPA on testicular cells and spermatozoa is associated with an increased production of reactive oxygen species, most of the pharmacological approaches are based on the use of natural or synthetic antioxidants.
