Most patients who developed these infections were taking concomitant immunosuppressants such as methotrexate or corticosteroids. Discontinue SIMPONI ARIA if a patient develops a serious infection. Reported infections with TNF blockers, of which SIMPONI ARIA is a member, include: Active tuberculosis, including reactivation of latent tuberculosis. Patients with tuberculosis have frequently presented with disseminated or extrapulmonary disease. Test patients for latent tuberculosis before SIMPONI ARIA use and during therapy. Initiate treatment for latent tuberculosis prior to SIMPONI ARIA use. Invasive fungal infections including histoplasmosis, coccidioidomycosis, candidiasis, aspergillosis, blastomycosis and pneumocystosis. Patients with histoplasmosis or other invasive fungal infections may present with disseminated, rather than localized, disease. Antigen and antibody testing for histoplasmosis may be negative in some patients with active infection. Consider empiric antifungal therapy in patients at risk for invasive fungal infections who develop severe systemic illness. Bacterial, viral, and other infections due to opportunistic pathogens, including Legionella and Listeria. Consider the risks and benefits of treatment with SIMPONI ARIA prior to initiating therapy in patients with chronic or recurrent infection. Monitor patients closely for the development of signs and symptoms of infection during and after treatment with SIMPONI ARIA, including the possible development of tuberculosis in patients who tested negative for latent tuberculosis infection prior to initiating therapy [see Warnings and Precautions (5.1)]. MALIGNANCY Lymphoma and other malignancies, some fatal, have been reported in children and adolescent patients treated with TNF-blockers, of which SIMPONI ARIA is a member [see Warnings and Precautions (5.2)]. 3 Reference ID: 4678119 1 INDICATIONS AND USAGE 1.1 Rheumatoid Arthritis (RA) SIMPONI ARIA, in combination with methotrexate (MTX), is indicated for the treatment of adult patients with moderately to severely active rheumatoid arthritis. 1.2 Psoriatic Arthritis (PsA) SIMPONI ARIA is indicated for the treatment of active psoriatic arthritis in patients 2 years of age and older. 1.3 Ankylosing Spondylitis (AS) SIMPONI ARIA is indicated for the treatment of adult patients with active ankylosing spondylitis. 1.4 Polyarticular Juvenile Idiopathic Arthritis (pJIA) SIMPONI ARIA is indicated for the treatment of active polyarticular juvenile idiopathic arthritis (pJIA) in patients 2 years of age and older.

FOR RESEARCH USE ONLY! TM BioSim anti-Ipilimumab (Yervoy®) (Human) ELISA Kit rev 07/18 (Catalog # E4398-100, 100 assays, Store at 4°C) I. Introduction: Ipilimumab (Yervoy®) is a fully human IgG1κ antibody that binds to CTLA-4 (cytotoxic T lymphocyteassociated antigen 4), a molecule on T- cells that is indicated for unresectable or metastatic melanoma. The absence or presence of CTLA-4 can augment or suppress the immune system's T-cell response in fighting disease. Ipilimumab is designed to block the activity of CTLA-4, thereby sustaining an active immune response in its attack on cancer cells. The proposed mechanism of action is indirect, and may be through T-cell - mediated anti-tumor immune responses. However, some patients develop unwanted immunogenicity, which leads to production of anti-drug-antibodies (ADAs) inactivating the therapeutic effects of the treatment and, in rare cases, inducing adverse effects. BioVision’s BioSimTM anti-Ipilimumab ELISA kit is designed to detect antibody against Ipilimumab with high specificity and sensitivity in biological matrices. II. Application: This ELISA kit is used for in vitro qualitative determination of antibody against Ipilimumab in serum and plasma Cross Reactivity: Ipilimumab (Yervoy®) infusion camouflages/masks the presence of antibody to Ipilimumab (ATP) in serum/plasma samples. Therefore, blood sampling time is critical for detection of ATP. It is convenient to obtain blood sample just before the infusion or at least 2 weeks after the infusion of Ipilimumab. III. Notes • There are several brands of targeted immune modulators on the market. There is a lack of reliable evidence that any one brand of targeted immune modulator is superior to other brands for medically necessary indications. Enbrel (etanercept), Humira (adalimumab), Remicade (infliximab), Simponi (golimumab), Simponi Aria (golimumab intravneous), and Stelara (ustekinumab) brands of targeted immune modulators ("least cost brands of targeted immune modulators") are less costly to the plan.

It impairs patients’ quality of life enormously. It has been recognized not only as a skin disease but as a systemic disease, since it also causes arthritis (psoriatic arthritis) and mental disorders. Furthermore, an association with cardiovascular events is indicated. With the advent of biologics, treatment of psoriasis dramatically changed due to its high efficacy and tolerable safety. A variety of biologic agents are available for the treatment of psoriasis nowadays. However, characteristics such as rapidity of onset, long-term efficacy, safety profile, and effects on comorbidities are different. Better understanding of those characteristic leads to the right choice for individual patients, resulting in higher persistence, longer drug survival, higher patient satisfaction, and minimizing the disease impact of psoriasis. In this paper, we focus on the efficacy and safety profile of biologics in psoriasis patients, including plaque psoriasis and psoriatic arthritis. In addition, we discuss the impact of biologics on comorbidities caused by psoriasis. Keywords: biologics; psoriasis; psoriatic arthritis; tumor necrosis factor-α; inteleukin-23; interluekin-17 1.

  • Golimumab: a New Anti-TNF Agent on the Horizon for Inflammatory Arthritis
DRUG EVALUATION Golimumab: a new anti-TNF agent on the horizon for infl ammatory arthritis Rheumatoid arthritis (RA), psoriatic arthritis, and ankylosing spondylitis are highly prevalent infl ammatory arthritides. Various conventional therapies are currently in use, and biologic therapies have also been introduced into the treatment armamentarium. Despite the remarkable effi cacy of the anti-tumor necrosis factor (TNF) agents, there still remains an unmet need for newer anti-TNF agents. A substantial number of patients do not respond to an initial anti-TNF agent, but may respond to a second one. The modes of administration of currently marketed anti-TNF agents are not particularly convenient and risk-free for all patients. An anti-TNF agent with a better safety profi le is also highly desirable. Golimumab (CNTO-148) is a high affi nity, human monoclonal antibody to TNF-α. The various ongoing trials for golimumab have shown promising results in terms of effi cacy and safety in methotrexate-naive and -resistant patients with RA, as well as in patients with RA who were previously treated with other anti-TNF agents. In addition, the effi cacy of golimumab in psoriatic arthritis and ankylosing spondylitis has also been demonstrated.
