The authors showed that 2AR can form a heteromeric complex with metabotropic glutamate receptor 2 (mGluR2) — which is a Gi protein-coupled receptor — and that this interaction can enhance glutamate-induced Gi signalling and reduce serotonin-induced Gq signalling. The intracellular Gi and Gq signalling balance is predictive of the anti- or pro-psychotic activity of drugs targeting 2AR and mGluR2, providing a potential new metric to improve current antipsychotic therapies. KAVA ( Piper methysticum ) - A REVIEW The Kava plant (Piper methysticum) is a robust, well-branching and erect perennial shrub belonging to the pepper family (Piperaceae). The botanical origin remains unknown, although it is likely that early Polynesian explorers brought the plant with them from island to island. Numerous varieties of Kava exist, and today it is widely cultivated in several Pacific Island countries both for local use as well as the rapidly growing demand for pharmaceutical preparations. The dried rhizomes (roots) are normally used. The first description to the western world of the ceremonial use of an intoxicating beverage prepared from Kava was made by Captain James Cook following his Pacific voyage in 1768. The drink, prepared as an infusion in an elaborate manner after first chewing the root, is consumed on formal occasions or meetings of village elders and chiefs, as well as in reconciling with enemies and on a more social basis. It remains an important social custom in many Pacific Island countries today. Most of the islands of the Pacific possessed Kava prior to European contact, particularly those encompassed by Polynesia, Melanesia and Micronesia. After drinking the Kava beverage a pleasantly relaxed and sociable state develops, after which a deep and restful sleep occurs. -

Origin and geographic distribution Datura stramonium is native to the Americas and has been introduced in many tropical, subtropical and even temperate regions. It is a naturalized weed in many African countries, but is probably seriously under-reported. Uses Datura stramonium and Datura metel L. have largely similar medicinal uses throughout the world. The most widely known use of Datura stramonium and of other Datura species is for relieving asthma, cough, tuberculosis and bronchitis by smoking the dried leaves, roots or flowers. ‘Asthma cigarettes’ have been shown to be very effective in some cases, but in other cases they had little or no effect. Cigarettes made with the leaves are also used to treat Parkinson’s disease. A decoction or infusion of leaves is given as a sedative to mental and schizophrenic patients. The leaves are applied as a dressing to cure rheumatic pain, swellings, wounds, gout, burns, ingrown toe-nails, fungal infections, tumours and ulcers. Dried pulverized leaves are dusted on wounds or applied after mixing the powder with fat or Vaseline. In DR Congo pounded fresh root and fresh leaves are soaked in water and the liquid is given in enema as an abortifacient. -

This study was designed to examine the resistance and virulence of poultryE. coli strains in vitro and in vivo via antibiotic susceptibility, biofilm formation and adhesion, and invasion and intracellular survivability assays in Caco-2 and Raw 264.7 cell lines as well as the determination of the median lethal dose in two-day old chickens. A clinical pathogenic multidrug-resistant isolate, E. coli 381, isolated from broilers, was found to be highly virulent in cell culture and 1000-fold more virulent in a chicken model than other strains; accordingly, the isolate was subsequently selected for transcriptome analysis. The comparative gene expression profile of MDRE. coli 381 and the reference human strain E. coli ATCC 25922 was completed with Illumina HiSeq. 2500 transcriptome analysis. Differential gene expression analysis indicates that there are multiple pathways involved in the resistance and virulence of this highly virulent strain. The results garnered from this study provide critical information about the highly virulent MDR E. coli strain of poultry origin and warrant further investigation due to its significant threat to public health. Escherichia coli is a Gram-negative bacterium that displays a wide range of genomic diversity. -

This study aims to examine whether administration of clonazepam (CZP; 1 mg/kg/day) in 7–11-day-old rats affects Gama aminobutyric acid (GABA)ergic receptors in both the short and long terms. Using RT-PCR and quantitative autoradiography, we examined the expression of the selected GABAA receptor subunits (α1, α2, α4, γ2, and δ) and the GABAB B2 subunit, and GABAA, benzodiazepine, and GABAB receptor binding 48 h, 1 week, and 2 months after treatment discontinuation. Within one week after CZP cessation, the expression of the α2 subunit was upregulated, whereas that of the δ subunit was downregulated in both the hippocampus and cortex.
