Afin de caractériser les mécanismes sous-jacents l’action de la DBS, et plus particulièrement, la possible implication du système glial, les effets de la stimulation du cortex préfrontal infralimbique sur plusieurs marqueurs précliniques de la réponse antidépressive ont été évalués chez le rat. Ce travail de thèse, en utilisant des approches électrophysiologiques, immunohistochimiques et comportementales, montre que la DBS aigue (130 Hz, 150 μA) induit des comportements pseudo- antidépresseurs (évalués dans le test de nage forcée) qui sont associés à une augmentation de l’activité des neurones 5-HT du raphé dorsal et de la neurogenèse du gyrus denté. De plus, la DBS aigue est capable de renverser les effets du stress sur la métaplasticité synaptique hippocampique. Par ailleurs, la DBS à plus faible intensité

Despite significant advancements in the devel- opment of new drugs with more acceptable side-effect profiles, patients with schizophrenia or major depression experi- ence substantial disability and burden of disease. The present review discusses the usefulness of partial dopamine D2/serotonin 5-HT1A receptors agonists in the treatment of schizophrenia, major depression and bipolar disorder as well as in Parkinson’s disease. Partial agonists can behave as modulators since their intrinsic activity or efficacy of a partial ago- nist depends on the target receptor population and the local concentrations of the natural neurotransmitter. Thus, these drugs may restore adequate neurotransmission while inducing less side effects. In schizophrenia, partial DA D2/5-HT1A receptor agonists (like aripiprazole or bifeprunox), by stabilizing DA system via a preferential reduction of phasic DA re- lease, reduce side effects i.e. extrapyramidal symptoms and improve cognition by acting on 5-HT1A receptors. Aripipra- zole appears also as a promising agent for the treatment of depression since it potentiates the effect of SSRIs in resistant treatment depression. Concerning bipolar disorders aripiprazole may have only a benefit effect in the treatment of manic episodes.

Ibarra3, Rubicelia Vargas4, Elisabeth Winnberg5 & Ana Martínez6* Dopamine is an important neurotransmitter that plays a key role in a wide range of both locomotive and cognitive functions in humans. Disturbances on the dopaminergic system cause, among others, psychosis, Parkinson’s disease and Huntington’s disease. Antipsychotics are drugs that interact primarily with the dopamine receptors and are thus important for the control of psychosis and related disorders. These drugs function as agonists or antagonists and are classifed as such in the literature. However, there is still much to learn about the underlying mechanism of action of these drugs. The goal of this investigation is to analyze the intrinsic chemical reactivity, more specifcally, the electron donor–acceptor capacity of 217 molecules used as dopaminergic substances, particularly focusing on drugs used to treat psychosis. We analyzed 86 molecules categorized as agonists and 131 molecules classifed as antagonists, applying Density Functional Theory calculations. Results show that most of the agonists are electron donors, as is dopamine, whereas most of the antagonists are electron acceptors. Therefore, a new characterization based on the electron transfer capacity is proposed in this study. This new classifcation can guide the clinical decision‑making process based on the physiopathological knowledge of the dopaminergic diseases. During the second half of the last century, a movement referred to as the third revolution in psychiatry emerged, directly related to the development of new antipsychotic drugs for the treatment of psychosis.
