on the evidence for the use of nutraceuticals for the prevention and treatment of important disease states. The use of nutraceuticals is by now an established complementary therapy, with a few being used as if they were conventional pharmaceuticals, and a number of others being purchased and used by the general public as self- medication. Although nutraceuticals are constantly discussed in the media and freely available, comprehensive knowledge concerning their activities, mode of action and safety is not yet widely available. The main aim of this book was, therefore, to explore, discuss and possibly substantiate claims that a number of nutraceuticals can actually treat or prevent the underlying causes of disease. Consumers clearly hope that they will benefit from intake of these products either through long-term use or via a conscious decision to change lifestyle. That is why I have decided here, unlike a number of other books on the subject, to organise the information according to disease states. In addition, any evidence quoted should be of under- standable quality. The inclusion of directly attributable information sources was considered vital for this, producing a usable text for both patient, consumer and healthcare practitioners wanting to find further details by checking the original publications. Wherever possible, I have tried to focus on clinical and human research, although results from animal studies are included, particularly for the chapter on animal health, but also when there are major findings or there are insufficient human data. Previous books have either focused on the individual chemical entities and their applications, or consist of information in therapeutic areas which is not directly attributed to published data, hence not allowing easy access to original material. In contrast, this text aims to cover a wide range of nutraceuticals, specifically focusing on their use in specific therapeutic categories.     Chapter Medicinal Properties of Selected Asparagus Species: A Review Polo-Ma-Abiele Hildah Mfengwana and Samson Sitheni Mashele Abstract Asparagus species are naturally distributed along Asia, Africa, and Europe and are known to have numerous biological properties. This review article was aimed to provide an organized summary of current studies on the traditional uses, phy- tochemistry, and pharmacological and toxicological studies of Asparagus laricinus Burch., Asparagus africanus Lam., Asparagus officinalis L., Asparagus racemosus Willd., and Asparagus densiflorus (Kunth) Jessop to attain and establish new insights for further researches. Information used in this review was obtained from electronic database including PubMed central, Google scholars, Science direct, Scopus, and Sabinet. Based on the present findings, the existing literature still presents some breaches about the mechanism of action of various constituents of these plants, and their relation to other plant compounds in poly-herbal formulations, as well as their long-term use and safety. More in-depth studies are still needed for active compounds and biological activities of Asparagus laricinus, Asparagus africanus, and Asparagus densiflorus. Therefore, innumerable opportunities and possibilities for investigation are still available in novel areas of these plants for future research stud¬ies. It can be concluded that all selected Asparagus species have tremendous potential to improve human health and the pharmacological activities of these plants can be attributed to bioactive phytochemicals they possess. Isolation and identification of phenolic compounds of the chloroform and n-butanol fractions were performed with Dionex HPLC-DAD system with water-methanol gradients in 4 different wave lengths (235 nm, 254 nm, 280 nm and 340 nm), using online UV and LC-MS as described previously. 9-OH-pinoresinol which is a lignan with anticancer activity was dominated in the chloroform fraction, whereas mainly flavonoid glycosides like quercetin-3-O-galactoside, apigenin-7-O-rhamnoside with anti-inflammatory effect were detected in the n-butanol fraction. Additionally, 9-OH-pinoresinol was also found in the n-butanol fraction. Anticancer tests were conducted in leukemia mouse lymphoma cells L5178Y at a concentration of 10 μg/ml of test compound. Crude ethanol extract of S. salicifolia reduced the growth of leukemia mouse lymphoma cells L5178Y to 23.8%. Bisphenol A (BPA) is a synthetic chemical used in the synthesis of polycarbonate plastics and epoxy resins. Endocrine disrupting properties of both genistein and BPA have been well established by various laboratories. Because the adverse biological effects caused by genistein and BPA are similar, and may include common co-exposure scenarios in the general population such as in the consumption of a soy-milk latte from a polycarbonate plastic coffee mug, analysis of the perturbation of the metabolism via glucuronidation of genistein in the presence of BPA has been assessed. Human and rat liver microsomes were exposed to varying doses of genistein (0 to 250 μM) in the absence (0 μM) or presence (25 μM) of BPA. Treatment with 25 μM BPA caused non-competitive inhibition of the glucuronidation of genistein in human liver microsomes with a Ki value of 58.7 μM, represented by a decrease in Vmax from 0.93 ± 0.10 nmol/min/mg in the ii absence of BPA to 0.62 ± 0.05 nmol/min/mg in the presence of BPA, and a negligible change in Km values between treatment groups.

Naturforsch. 60c, 139Ð142 (2005); received November 9/December 8, 2004 There is a wealth of evidence that hepatic stellate cells (HSCs) orchestrate most of the important events in liver fibrogenesis. After liver injury, HSCs become activated to a profi- brogenic myofibroblastic phenotype and can regulate net deposition of collagens and other matrix proteins in the liver. The proliferation of HSCs is mainly stimulated by the platelet- derived growth factor (PDGF). In this study, some compounds from natural resources have been tested for their activity to inhibit PDGF-driven proliferative activity of rat HSCs. Api- genin, quercetin, genistein, daidzin, and biochanin A exhibited > 75% inhibitory activity against HSC-T6. It was found that, γ-linolenic (γ-Ln), eicosapentanoic (EPA) and α- linolenic (α-Ln) acids showed a high inhibitory effect on proliferation of rat HSCs at 50 nmol/l. Cholest-4-ene-3,6-dione and stigmastone-4-en-3,6-dione are the most active steroids with in- hibitory activities > 80% and this is most likely due to the presence of the 4-en-3,6-dione moiety in both compounds.

A total of 25 isoflavones, 13 saponins and 3 flavones were identified by co-chromatography of sample extract with authentic standards and comparing the retention time, UV spectra, characteristic molecular ions and fragment ions with those of authentic standards, or tentatively identified by MS/MS determination along with Mass Fragment software. Moreover, the method was validated for the simultaneous quantification of 29 components. The samples from two Pueraria flowers significantly differed in the quality and quantity of isoflavones, saponins and flavones, which allows the possibility of showing their chemical distinctness, and may be useful in their standardiza- tion and quality control. Dataset obtained from UPLC-MS was processed with principal component analysis (PCA) and orthogonal partial least squared discriminant analysis (OPLS-DA) to holistically compare the dif- ference between both Pueraria flowers.

Arnold Abstract Dehydroepiandrosterone (DHEA) is used as a dietary supplement and can be metabolized to androgens and/or estrogens in the prostate. We investigated the hypothesis that DHEA metabolism may be increased in a reactive prostate stroma environment in the presence of proinflammatory cytokinessuchastransforminggrowth factor β1(TGFβ1), and further, whether red clover extract, which contains a variety of compounds including isoflavones, can reverse this effect. LAPC-4 prostate cancer cells were grown in coculture with prostate stromal cells (6S) and treated with DHEA +/− TGFβ1 or interleukin-6. Prostate-specific anti- gen (PSA) expression and testosterone secretion in LAPC-4/6S cocultures were compared with those in monocultured epithelial and stromal cells by real-time PCR and/or ELISA. Combined administration of TGFβ1 + DHEA to cocultures increased PSA protein secretion two to four times, and PSA gene expression up to 50-fold. DHEA + TGFβ1 also increased coculture production of testosterone over DHEA treatment alone. Red clover isoflavone treatment led to a dose-dependent decrease in PSA protein and gene expression and tes- tosterone metabolism induced by TGFβ1 + DHEA in prostate LAPC-4/6S cocultures. In this coculture model of endocrine-immune-paracrine interactionsin the prostate,TGF β1 greatly increased stromal-mediated DHEA effects on testosterone production and epithelial cell PSA production, whereas red clover isoflavones reversed these effects.

Knudsen, Ph.D. Abstract Due to its pivotal role in prostatic growth and survival, the androgen receptor (AR) is the primary target of disseminated prostate cancer (CaP), as achieved via androgen deprivation therapy (ADT). Unfortunately, ADT is circumvented by restoration of AR activity, resulting in ADT resistant tumors for which there is no alternate treatment option. Through multiple mechanisms, reactivation of the AR specifically underlies the progression to therapy resistant tumors. The environmentally prevalent endocrine disrupting compound (EDC), bisphenol A (BPA) is able to activate specific somatically mutated ARs commonly found in CaP, resulting in androgen-independent proliferation of CaP cells. To directly assess the effect of BPA on ADT, we used an in vivo xenograft model of CaP that expresses a BPA-sensitive mutant AR, and mimics standard human cytotoxic response to ADT, followed by subsequent tumor re-growth. When tumor- bearing animals were exposed to environmentally relevant levels of BPA during ADT, the tumors failed therapy more rapidly (compared to placebo controls), with AR re- activation and concomitant increased tumor cell proliferation. These data suggest that environmentally relevant exposure to EDCs may reduce the efficacy of mainline ADT for CaP.
