The genetics and evolution of iridescent structural colour in Heliconius butterflies Melanie N. Brien A thesis submitted in partial fulfilment of the requirements for the degree of Doctor of Philosophy The University of Sheffield Faculty of Science Department of Animal & Plant Sciences Submission Date August 2019 1 2 Abstract The study of colouration has been essential in developing key concepts in evolutionary biology. The Heliconius butterflies are well-studied for their diverse aposematic and mimetic colour patterns, and these pigment colour patterns are largely controlled by a small number of homologous genes. Some Heliconius species also produce bright, highly reflective structural colours, but unlike pigment colour, little is known about the genetic basis of structural colouration in any species. In this thesis, I aim to explore the genetic basis of iridescent structural colour in two mimetic species, and investigate its adaptive function. Using experimental crosses between iridescent and non-iridescent subspecies of Heliconius erato and Heliconius melpomene, I show that iridescent colour is a quantitative trait by measuring colour variation in offspring. I then use a Quantitative Trait Locus (QTL) mapping approach to identify loci controlling the trait in the co-mimics, finding that the genetic basis is not the same in the two species. In H. erato, the colour is strongly sex-linked, while in H. melpomene, we find a large effect locus on chromosome 3, plus a number of putative small effect loci in each species. Therefore, iridescence in Heliconius is not an example of repeated gene reuse. I then show that both iridescent colour and pigment colour are sexually dimorphic in H. -
The Speciation History of Heliconius: Inferences from Multilocus DNA Sequence Data
The speciation history of Heliconius: inferences from multilocus DNA sequence data by Margarita Sofia Beltrán A thesis submitted for the degree of Doctor of Philosophy of the University of London September 2004 Department of Biology University College London 1 Abstract Heliconius butterflies, which contain many intermediate stages between local varieties, geographic races, and sympatric species, provide an excellent biological model to study evolution at the species boundary. Heliconius butterflies are warningly coloured and mimetic, and it has been shown that these traits can act as a form of reproductive isolation. I present a species-level phylogeny for this group based on 3834bp of mtDNA (COI, COII, 16S) and nuclear loci (Ef1α, dpp, ap, wg). Using these data I test the geographic mode of speciation in Heliconius and whether mimicry could drive speciation. I found little evidence for allopatric speciation. There are frequent shifts in colour pattern within and between sister species which have a positive and significant correlation with species diversity; this suggests that speciation is facilitated by the evolution of novel mimetic patterns. My data is also consistent with the idea that two major innovations in Heliconius, adult pollen feeding and pupal-mating, each evolved only once. By comparing gene genealogies from mtDNA and introns from nuclear Tpi and Mpi genes, I investigate recent speciation in two sister species pairs, H. erato/H. himera and H. melpomene/H. cydno. There is highly significant discordance between genealogies of the three loci, which suggests recent speciation with ongoing gene flow. Finally, I explore the phylogenetic relationships between races of H. melpomene using an AFLP band tightly linked to the Yb colour pattern locus (which determines the yellow bar in the hindwing). -

M., Universidade Federal do Rio de J a neiro, llha do Fundil.o, Rio de Janeiro ZC-32, Brazil INTRODUCTION The rare mimetic species Heliconius demeter Staudinger was the subject of a recent taxonomic revision (Turner, 1966), in which a new subspecies was described (H. d. beebei from Guyana) and the complicated synonymy of the other three subspecies (demeter, bouqueti Noldner, and eratosignis Joicey & Talbot) was summarized. In the most recent complete revision of the genus (Emsley, 1965), the species was placed near Hel. ricini (L.) in the sara-group. The unusually small wingspans of demeter and ricini had led many earlier authors to place them both in the genus Eueides, but they are in fact very distant from members of this part of the tribe. All of the known subspecies of demeter except eratosignis were illustrated in a recent compilation of photographs of little-known Heliconius (Turner, 1973); eratosignis was pictured as part of a mimetic group from Rondonia in southwestern Brazil (Brown, 1973); the types of this form (in the British Museum (Natural History)) are shown in Figure 1. -

//FS2/CUP/3-PAGINATION/SPDY/2-PROOFS/3B2/9780521883184C10.3D 177 [177–194] 19.9.2008 3:02PM CHAPTER TEN Rapid speciation, hybridization and adaptive radiation in the Heliconius melpomene group james mallet In 1998 it seemed clear that a pair of ‘sister species’ of tropical butterflies, Heliconius melpomene and Heliconius cydno persisted in sympatry in spite of occasional although regular hybridization. They speciated and today can coexist as a result of ecological divergence. An important mechanism in their speciation was the switch in colour pattern between different Mu¨ llerian mimicry rings, together with microhabitat and host-plant shifts, and assortative mating pro- duced as a side effect of the colour pattern differences. An international con- sortium of Heliconius geneticists has recently been investigating members of the cydno superspecies, which are in a sense the ‘sisters’ of one of the original ‘sister species’, cydno. Several of these locally endemic forms are now recognized as separate species in the eastern slopes of the Andes. These forms are probably most closely related to cydno, but in several cases bear virtually identical colour patterns to the local race of melpomene, very likely resulting from gene transfer from that species; they therefore can and sometimes do join the local mimicry ring with melpomene and its more distantly related co-mimic Heliconius erato. I detail how recent genetic studies, together with ecological and behavioural observations, suggest that the shared colour patterns are indeed due to hybrid- ization and transfer of mimicry adaptations between Heliconius species. These findings may have general applicability: rapidly diversifying lineages of both plants and animals may frequently share and exchange adaptive genetic variation. -

Entomol. 45.- A remarkable new subspecies of the Amazonian and Guianan Heliconius burneyi (Hübner) is described from the state of Sucre in northeastern Venezuela. Its wing pattern resembles that of the peculiar heliconiine Neruda metharme (Erichson), which is absent in Sucre. Considering that races of H. burneyi are variations on basically the same theme, a new wing pattern element for this species is recognized. Natural selection, shifting balance and relictual mimicry may explain its occurrence. Key Words: Aposematísm, evolución, Paria, taxonomy. RESUMEN ORELLANA AM. 2006. Una notable subespecie nueva de Heliconius Kluk del nororiente de Venezuela (Lepidoptera: Nymphalidae). Rev. perú. A preliminary checklist of over 800 identified species is given, and some of the factors contributing to this diversity are described. The tropical rain forest in this area is being rapidly cleared for development and the creation of one or more inviolate biological preserves is urgently needed in order to save a living sample of the incredibly diverse fauna and flora for study by future generations. Rondonia, one of the newest states in western central Brazil, occupies some 93,840 square miles (243,044 sq km) in the southwestern part of the Amazon Basin of South America. This territory, which borders Bolivia to the south and west, was formerly a part of the state of Amazonas and until the last two decades, was primarily important to Brazil's economy only during the Amazon rubber boom, which collapsed in 1912. In 1943, the area was established as Guapore. -

We must stress however that comparisons of faunistic lists of different sites are always difficult to be made due to differences in area size, to divergence in classification, and to the differences of the operators techniques. Therefore it is often uneasy to deal with sets of data and to compare the results. None the less, surveys and comparisons of butterflies in selected sites of the Neotropical realm seem to be very popular nowadays (Lamas , 1983a, 1983b; Lamas et al., 1991; Raguso & Llorente , 1991; Austin et al., 1996; Balcazar , 1993). They are particularly aimed at gathering sets of data tor conservation purposes and at identifying hotspots of endemicity. Dramatic is the lacking of published long-term surveys on moths in limited areas of the Neotropics. Unexpectedly, no recent faunistic lists of Ecuadorian butterflies have appeared except that of Ma- quipucuna Reserve on the western side of the country (Raguso & G loster , 1996). Having observed butterflies, Saturniids and Hawkmoths for almost 15 years in Ecuador, as Clench (1979) suggests, the time is arrived to submit a survey of these taxa occuring in an Amazonian area of Ecuador. -

Although they are widespread throughout the mid-West, in this part of the country they are • • • considered a fugitive species-as the name implies, here today, and mysteriously gone tomorrow. Habitat requirements seem to dictate the length of their stay but are poorly understood. In April and May 2011 , thanks to Irving Finkelstein's years of experience and his gracious generosity, Sara Bright and I were able to explore the habitat and lifestyle of Gorgone Checkerspots at Cooper's Furnace Day Use Area near Clarksville, Georgia. Cooper's Furnace Day Use Area is located on the banks of the Etowah River, near Altoona Dam. Currently operated by the U.S. Army Corps of Engineers, it served as an antebellum industrial center and is on the National Registry of Historic Places. Its trails wind through a lovely mixed pine and hardwood forest complete with mountain overlooks, picturesque streams, and a beaver pond. Irving has a long history with this site. In the mid-1970s when he decided to explore natural areas within a 50-mile radius of his new home in Atlanta, one reconnaissance mission took him to the Cooper's Furnace area where he was delighted to discover a colony of Gorgone Checkerspots. -

Notes on systematics are given and data on bionomics of the species mostly from field experience in the last 15 years. The major taxonomic changes are as follows: Altinote erinome mathani (Oberthür, 1917) stat. rev. et comb, nov., Altinote hilaris desmiala (Jordan, 1913) stat. rev. et comb. nov. The nominal taxa laeta Oberthür, 1917 and amphilecta Jordan, 1913, are sunk as junior synonyms of diversa Jordan, 1913 and aereta Jordan, 1913, respectively. These taxa are considered to be the two Ecuadorian subspecies of Altinote stratonice (La- treille, 1817). Altinote erebia (Oberthür, 1917) is assumed to be a species distinct from Altinote tenebrosa (Hewitson, 1868).
