ABSTRAK Isolasi senyawa fenolik yang terdapat pada daun tumbuhan kayu hitam (Diospyros celebica Bakh.) (Ebenaceae) telah dilakukan dengan ekstraksi maserasi menggunakan pelarut methanol. Ekstak pekat metanol dilarutkan dengan aquadest dan dipartisi menggunakan pelarut etil asetat lalu dilarutkan dengan pelarut methanol dan dipartisi dengan n-heksana . Kemudian di analisa dengan kromatografi lapis tipis dan dipisahkan dengan kromatografi kolom dengan eluen Kloroform : Metanol (90:10) v/v; (80:20) v/v; (70:30) v/v; (60:40) v/v; (50:50) v/v. Isolat dimurnikan dengan kromtografi lapis tipis preparative dan di kristalisasi , diperoleh kristal berwarna kuning sebanyak 7,5 mg, dengan harga Rf = 0.45. Uji kemurnian senyawa yang diperoleh dilakukan dengan kromatografi lapis tipis menunjukkan noda tunggal menggunakan eluen kloroform : etil asetat ( 50:50) v/v, n-heksana : etil asetat (80:20) v/v , selanjutnya senyawa yang diperoleh dianalisis dengan Spektofotometer UV-Visible, Spektrofotometer Inframerah (FT-IR), dan Spektrofotometer resonansi magnetik inti proton (1H-NMR). Dari interpretasi data spektroskopi, diduga bahwa senyawa hasil isolasi yang diperoleh adalah senyawa fenolik metil galat golongan asam fenolat. Kata Kunci : Daun Kayu Hitam (diospyros delebica Bakh.), Fenolik, Metil Galat, iii Universitas Sumatera Utara ISOLATION AND IDENTIFICATION OF PHENOLIC COMPOUNDS FROM KAYU HITAM LEAVES (Diospyros celebica Bakh.) ABSTRACT Isolation of phenolic compounds from kayu hitam leaves (Diospyros Celebica Bakh.) has been done by maceration technique with methanol solvent.The concentrated exstract of methanol dissolved with aquadest and partition exstrakted with ethylacetate. The concentrated extract of ethyl acetate dissolved with methanol solvent, and partition with n-hexane. Then Thin layer chromatograpy was analyzed before coloumn chromatography and the eluen obtained that is suitable for separation is chloroform : methanol 90:10, 80:20, 70:30,60:40,50:50 (v/v) as mobile phase.The compounds was purified with a thin layer preparative and cristalitation yielding yellow pasta with weigh 7,5 mg with Rf = 0,45. The test purity of compounds is done by thin layer chromathography show sigle spot with eluen chloroform : ethyl acetate (50:50) v/v ang n-hexane : ethyl acetate ( 80:20) v/v . the next,compound was further indified by Spectrophotometer ultraviolet Visible (UV-Vis), Fourier Transform Infra Red Spectrophotometer (FT-IR) and Proton Nuclear Magnetic Resonancy Spectrophotometer (1H-NMR) was estimated as a phenolic methyl gallate. According to the report of the Food and Agriculture Organization of the United Nation (FAO), approximately 13 million hectors of global forests have been lost annually due to forest land conversion to other land uses, forest fires and natural disasters, while reforestation and natural regeneration account for an increase of approx. 7.8 million hectors of forest cover. This means the net loss of global forest is estimated at 5.2 million hectors. Adverse impacts of forest conversion to farmland can be minimized as far as the land is properly used and managed in a sustainable manner. However, in some cases, problem soils are exposed and abandoned as degraded land. Deforestation by mining is a big issue these years. Problem soils such as strong acid soils and/or too much heavy metal soils appear at the ex-mining areas. In some cases it is too difficult to reforestate.

Due to this condition, IUCN in 1998 listing this species into vulnerable species. Conservation effort and breeding programs are among the urgent activities needed to prevent extinction. Genetic characteristics of the species and population is needed as basic consideration in formulating appropriate conservation strategy for the species. Our research was conducted to determined ebony genetic diversity at parental and offspring stages. Simple sequence repeat (SSR) markers were used to analyze 164 individuals (92 trees and 72 seedlings). Molecular data were processed using Gene Alexand DARw into analyze allelic frequencies and dendrograms clustering.

Taxonomy and nomenclature Family: Ebenaceae Synonym: Sirium myrtifolium L., Santalum ovatum R. Br., Santalum myrtifolium (L) Roxb. Vernacular/common name: Trade name ebony used for all dark-wooded Diospyros species throughout the tropics. Ebony is thus locally used for different lo- cal species. Other local names for D. celebica in In- donesia are Kayu Hitam, Toe (Donggala, Poso and Ma- nado), Limara (Luwu), Sora (Malili), and Ayu Maitong (Parigi). Distribution and habitat Diospyros celebica Bakh. is endemic to the eastern part of Indonesia, occuring naturally in Sulawesi and possibly the Maluku. In Sulawesi it grows particularly in North Sulawesi (Minahasa, Bolaang Mongondow), Central Sulawesi (Parigi, Poso, Donggala, Toli-toli, Kolonodale, and Luwuk), and South Sulawesi (Maros, Barru, Luwu, and Mamuju).     ANATOMICAL AND PHYSICAL PROPERTIESOF BISBUL WOOD (Diospyros blancoi A.DC.) Krisdianto and Abdurachman' ABSTRACT Ebony (Diospyros sp.) is a heavy hardwood that is popularly known as blackwood. Diospyros consists of over 300 species spread throughout tropics area and about 100 species occur in Indonesia. Bisbul wood (Diospyrosblan coi A.DC.) is one species of streaked ebony that is locally known as 'buah mentega'. The anatomical and physical properties of bisbul were studied to collect information for wood identification and to support appropriate use of the timber. Anatomical properties were studied from microtome sectioned samples, which have been coloured by safranin and mounted by entellan, while fiber dimensions were studied from macerated samples. Physical properties of bisbul wood studied include moisture content, density and percentage of volumetric shrinkage. Moisture content and density were studied from 20 x 20 x 20 mm samples based on wet and oven dry condition. Volumetric shrinkage was measured from dimension changes in radial, tangential and longitudinal shrinkage of 20 x 20 x 40 mm samples. The samples were measured in wet and oven dry conditions. The main anatomical characteristics to identify bisbul wood were black wood with pinkish streaked, heavy and very hard, very fine texture, even, lustrous surface and glossy, distinct growth ring, small size of vessels, apotracheal parenchyma forming reticulate pattern. The average moisture content was 59 .86 ± 2.84%, the density average was 0.74 ± 0.04 gr/cm' and volumetric shrinkage was 10.41±0.70%. The higher the stem, the more moisture content and the lower the density will be. Sapwood density was lower and had more moisture content than heartwood.

The Enrekang Botanic Garden is newly established in Kabupaten Enrekang, South Sulawesi, Indonesia, to document and conserve the diversity of plants from the Wallacea Region. The new botanic garden is a collaborative venture since 2005, in which Kebun Raya Bogor is assisting the local authorities of Sulawesi Selatan to form a development plan and establish the garden’s living collections. The garden has developed nursery facilities, an irrigation system, and road access. A total of 4601 living plants have been planted, representing 36 families, 156 genera and 232 species. Keywords. It has revealed that depending on the natural origin of the wood species studied and harmonized with the ethnobotanical and ethnomedicinal knowledge, the extractives derived from the woody plants have given valuable heritages in the fields of medicines and phar- macology. The families of the bioactive compounds found in the extracts mainly consisted of flavonoids, stilbenes, stilbenoids, lignans, tannins, simple phenols, terpenes, terpenoids, alkaloids, quinones, and saponins. In addition, biological or pharmacological activities of the extracts/isolated phytochemicals were recorded to have antioxidant, antimicrobial, antifungal, anti-inflammatory, anti-diabetes, anti-dysentery, anticancer, analgesic, anti-malaria, and anti-Alzheimer activities.
