Make sure you are following Procedures on p. 90-91 using 1% amylase.
Be extremely careful with pipettes - what would happen if you double-dipped into pH 5 and then 7 without rinsing/changing pipettes?
Begin by analyzing enzyme treated with pH 7 - if this shows no activity, there is a serious reagent or equipment problem and you need to start over.
If pH 7 worked, proceed through 6.5, 7.5, 8, 5.5, and 5 in that order - one at a time to avoid double-dipping and mixing things up.

Make sure you are following Procedures on p. 94-95 using 1% amylase.
Make sure you leave all tubes at the correct temperature while treating AND testing.
