Laboratory Procedures

5’-Iodoresinferatoxin (I-RTX) is supplied as a solid. A stock solution may be made by dissolving the I-RTX in the solvent of choice. I-RTX is soluble in organic solvents such as and DMSO, which should be purged with an inert gas. I-RTX is sparingly soluble in aqueous solutions. To enhance aqueous solubility, dilute the organic solvent solution into aqueous buffers or isotonic saline. If performing biological experiments, ensure the residual amount of organic solvent is insignificant, since organic solvents may have physiological effects at low concentrations. We do not recommend storing the aqueous solution for more than one day.

Description

1 I-RTX is a potent and reversible antagonist of TRPV1 (Kd = 4.3 nM). However, it can also act as a partial TRPV1 at higher concentrations in vitro, inducing a biphasic response with EC50 values of 56.7 nM and 9.9 μM.2 I-RTX blocks activation of TRPV1 by both in vitro and in vivo.1,3,4 It also blocks neurogenic pain induced by inflammatory mediators.5,6
