More than 30 species have been isolated but only N. fowleri has been associated with human disease. N. fowleri causes primary amoebic meningoencephalitis (PAM), an acute, often fulminant infection of CNS. Here we report a rare and first case of PAM in an immunocompetent elderly patient from this part of the country. Amoeboid and flagellate forms of N. fowleri were detected in the direct microscopic examination of CSF and confirmed by flagellation test in distilled water, demonstrating plaques /clear areas on 1.5% non nutrient agar and its survival at 42°C. MACIVER Biomedical Sciences, Edinburgh Medical School, University of Edinburgh, Scotland, UK Abstract. We report the isolation of a new species of Allovahlkampfia, a small cyst-forming heterolobosean soil amoeba. Phylogenetic analysis of the 18S rDNA and the internal transcribed spacers indicates that Allovahlkampfia is more closely related to the acrasids than to other heterolobosean groups and indicates that the new strain (GF1) groups with Allovahlkampfia tibetiensisand A. nederlandiensis despite being significantly smaller than these and any other described Allovahlkampfia species. GF1 forms aggregated cyst masses similar to the early stages of Acrasis sorocarp development, in agreement with the view that it shares ancestry with the acrasids. Time-lapse video mi- croscopy reveals that trophozoites are attracted to individuals that have already begun to encyst or that have formed cysts. Although some members of the genus are known to be pathogenic the strain GF1 does not grow above 28oC nor at elevated osmotic conditions, indicating that it is unlikely to be a pathogen. Memoirs of the Queensland Museum 45(1):1-164. Brisbane. ISSN 0079-8835. Published reports of protozoan species from Australian animals have been compiled into a host- parasite checklist, a parasite-host checklist and a cross-referenced bibliography. Protozoa listed include parasites, commensals and symbionts but free-living species have been excluded. Over 590 protozoan species are listed including amoebae, flagellates, ciliates and ‘sporozoa’ (the latter comprising apicomplexans, microsporans, myxozoans, haplosporidians and paramyxeans). Organisms are recorded in association with some 520 hosts including mammals, marsupials, birds, reptiles, amphibians, fish and invertebrates. Information has been abstracted from over 1,270 scientific publications predating 1999 and all records include taxonomic authorities, synonyms, common names, sites of infection within hosts and geographic locations. Protein secretion and encystation in Acanthamoeba Alvaro de Obeso Fernández del Valle Doctor of Philosophy The University of Edinburgh 2018 Abstract Free-living amoebae (FLA) are protists of ubiquitous distribution characterised by their changing morphology and their crawling movements. They have no common phylogenetic origin but can be found in most protist evolutionary branches. Acanthamoeba is a common FLA that can be found worldwide and is capable of infecting humans. The main disease is a life altering infection of the cornea named Acanthamoeba keratitis. Additionally, Acanthamoeba has a close relationship to bacteria. Acanthamoeba feeds on bacteria. At the same time, some bacteria have adapted to survive inside Acanthamoeba and use it as transport or protection to increase survival. When conditions are adverse, Acanthamoeba is capable of differentiating into a protective cyst. -

SANIT., M.SC. 3 RESUMEN Los protozoarios patógenos Cryptosporidium parvum y Giardia sp. han demostrado su infectividad e impacto negativo en la salud de miles de personas tanto en naciones industrializadas como en los países en desarrollo. La mayoría de los protozoarios por presentar una forma resistente a las condiciones ambientales, les permite la supervivencia a los tratamientos físico-químicos del agua para consumo humano. De igual forma, la aparición de nuevos patógenos demuestra la necesidad de desarrollar nuevos indicadores de calidad microbiológica que permitan ofrecer productos verdaderamente seguros en el agua para uso humano. El presente artículo es una revisión de literatura que señala el impacto de este riesgo microbiológico, asociado fundamentalmente con el consumo de aguas cuyos indicadores clásicos de contaminación microbiológica (coliformes fecales y Escherichia coli) en casi todos los casos cumplen con las normas vigentes. Parasitic protozoa transmission by drinking water SUMMARY The pathogenic protozoa Cryptosporidium sp. and Giardia sp. showed their infectivity and negative effects on the health of thousands of people in both developed and developing countries. Protozoa have ambient resistant stages that permit survived to physical and chemical treatment of drinking water. This risk is mainly related to the consumption of water whose microbiological quality indicators typically fulfill with current standards (fecal coliforms and Escherichia coli). -

More than 40 species have been characterized, but only Naegleria fowleri (N. fowleri) is known as a human pathogen causing primary amoebic meningoencephalitis (PAM), a fast progressing and mostly fatal disease of the central nervous system. Several studies report an involvement of phospholipases and other molecular factors, but the mechanisms involved in pathogenesis are still poorly understood. To gain a better understanding of the relationships within the genus of Naegleria and to investigate pathogenicity factors of N. fowleri, we characterized the genome of its closest non-pathogenic relative N. lovaniensis. Results: To gain insights into the taxonomy of Naegleria, we sequenced the genome of N. lovaniensis using long read sequencing technology. The assembly of the data resulted in a 30 Mb genome including the circular mitochondrial sequence. Unravelling the phylogenetic relationship using OrthoMCL protein clustering and maximum likelihood methods confirms the close relationship of N. lovaniensis and N. fowleri.
