Western Blot Flow Cytometry Detection of Human CD302/CLEC13A by Western Blot. Western Detection of CD302/CLEC13A in blot shows lysates of HL­60 human acute promyelocytic leukemia cell Human Monocytes by Flow line. PVDF Membrane was probed with 1 µg/mL of Sheep Anti­Human Cytometry. Human peripheral blood CD302/CLEC13A Antigen Affinity­purified Polyclonal Antibody monocytes were stained with Sheep (Catalog # AF6367) followed by HRP­conjugated Anti­Sheep IgG Anti­Human CD302/CLEC13A Affinity Secondary Antibody (Catalog # HAF016). A specific band was detected Purified Polyclonal Antibody for CD302/CLEC13A at approximately 30 kDa (as indicated). BACKGROUND CD302 (also DCL­1 and CLEC13A) is a 31­36 kDa member of the C­type domain family of proteins. It is expressed on cells involved in , including neutrophils, monocytes, dendritic cells and macrophages. CD302 is suggested to facilitate phagocytosis, and participate in . Mature human CD302 is a 210 amino acid (aa) type I transmembrane glycoprotein. It contains a 146 aa extracellular domain (ECD) (aa 23­168) plus a 43 aa cytoplasmic region. The ECD possesses a C­type lectin domain (aa 32­152) that apparently cannot bind carbohydrate. CD302 has also been detected in dendritic cells as the C­terminal portion of a 215 kDa fusion that involves DEC205. CD302 contributes aa 23­232 to the fusion protein. There is one potential alternative start site at Met63. Over aa 23­168, human CD302 shares 82% aa identity with mouse CD302.
