Western Blot Flow Cytometry Detection of Human CD302/CLEC13A by Western Blot. Western Detection of CD302/CLEC13A in blot shows lysates of HL60 human acute promyelocytic leukemia cell Human Monocytes by Flow line. PVDF Membrane was probed with 1 µg/mL of Sheep AntiHuman Cytometry. Human peripheral blood CD302/CLEC13A Antigen Affinitypurified Polyclonal Antibody monocytes were stained with Sheep (Catalog # AF6367) followed by HRPconjugated AntiSheep IgG AntiHuman CD302/CLEC13A Affinity Secondary Antibody (Catalog # HAF016). A specific band was detected Purified Polyclonal Antibody for CD302/CLEC13A at approximately 30 kDa (as indicated). This (Catalog # AF6367, filled histogram) experiment was conducted under reducing conditions and using or control antibody (Catalog # 5001 Immunoblot Buffer Group 1. BACKGROUND CD302 (also DCL1 and CLEC13A) is a 3136 kDa member of the Ctype lectin domain family of proteins. It is expressed on cells involved in phagocytosis, including neutrophils, monocytes, dendritic cells and macrophages. CD302 is suggested to facilitate phagocytosis, and participate in cell adhesion. Mature human CD302 is a 210 amino acid (aa) type I transmembrane glycoprotein. It contains a 146 aa extracellular domain (ECD) (aa 23168) plus a 43 aa cytoplasmic region. The ECD possesses a Ctype lectin domain (aa 32152) that apparently cannot bind carbohydrate. CD302 has also been detected in dendritic cells as the Cterminal portion of a 215 kDa fusion protein that involves DEC205. CD302 contributes aa 23232 to the fusion protein. There is one potential alternative start site at Met63. Over aa 23168, human CD302 shares 82% aa identity with mouse CD302.
