SERMs such as toremifene, raloxifene and tamoxifen suppressed the proliferation of prostate epithelial and stromal cells whereas anti-androgens did not. In prostate stromal cells, the transactivation activities of ER were enhanced by adding E2 and reduced remarkably by toremifene. The results indicate that the ER-mediated pathway plays a central role in the growth of normal prostate cells. One of the hypothesized in the development of the prostate gland and the pathogeneses of BPH is that the stromal hyperplasia is pathogenesis of prostatic diseases.1 Maternal estradiol, related to an enhanced estrogenic status after middle age, for example, stimulates squamous metaplasia within the with a decreased androgen level and an increased developing prostatic epithelium in a male offspring.2 conversion of adrenal androgens to estrogen by aroma- With regard to localized prostate cancer, expression tase.11 Recently, key studies on the role of estrogen in the levels of the estrogen receptor b (ERb) decline with pathogenesis of BPH have been carried out using normal increasing grade from prostatic intraepithelial neoplasia human prostate stromal and epithelial cells.
