BACKGROUND Introduction Speckle-type pox virus and zinc finger (POZ) protein, also known as HIB homolog 1 or Roadkill homolog 1, is encoded by the SPOP gene (also known as TEF2 or BTBD32) in human (Entrez Gene ID 8405). SPOP protein is a component of cullin-RING-based BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex and mediates recruitment of target proteins containing the SPOP-binding consensus (SBC) motif. The MATH domain is primarily involved in substrate recognition/binding. Known substrates with SBC motifs include Macro H2A, Puc (Puckered), Daxx (Death domain–associated protein), and Gli (Glioma-associated oncogene). Prostate and endometrial cancers are frequently identified with SPOP mutations in the meprin and TRAF (Tumor necrosis factor receptor-associated factor) homology (MATH) domain, which can potentially lead to altered substrate binding activities. The SPOP protein sequence is 100% conserved among human, murine, and rat (UniProt O43791, Q6ZWS8, and B2RYD9, respectively) species.
