Mouse GDF-5/BMP-14 Biotinylated Antibody Antigen Affinity-purified Polyclonal Goat IgG Catalog Number: BAF853 DESCRIPTION Species Reactivity Mouse Specificity Detects mouse GDF5/BMP14 in Western blots. In Western blot, approximately 20% crossreactivity with recombinant mouse (rm) GDF6 is observed and less than 1% crossreactivity with rmGDF1, rmGDF8, and rmGDF9 is observed. Source Polyclonal Goat IgG Purification Antigen Affinitypurified Immunogen E. coliderived recombinant mouse GDF5/BMP14 Ala376Arg495 Accession # P43027 Formulation Lyophilized from a 0.2 μm filtered solution in PBS with BSA as a carrier protein. BACKGROUND Growth Differentiation Factor 5 (GDF5), also known as cartilagederived morphogenetic protein 1 (CDMP1), is a member of the bone morphogenetic protein (BMP) family which belongs to the transforming growth factor β (TGFβ) superfamily. GDF5 is synthesized as a large precursor protein that consists of an Nterminal 19 amino acid (aa) signal sequence, a 362 aa pro region and a 120 aa Cterminal mature peptide. Mature GDF5 is a homodimeric protein which contains the characteristic seven conserved cysteine residues. GDF5, GDF6 and GDF7, which share 8086% identity, define a new subgroup within the BMP family. Like other TGFβ superfamily proteins, GDF5 is highly conserved across species. At the amino acid sequence level, mature human and mouse GDF5 are 98% identical. It has been reported that GDF5 has multiple functions including regulation of myogenesis, regulation of chondrogenesis, bone morphogenesis, and neuron differentiation and survival. GDF5 response is mediated by the formation of heterooligomeric complexes of type I (BMPRIB) and type II (BMPRII or Activin RII) sereine/threonine kinase receptors, and the activation of Smad proteins (Smad 1, 5, and 8).
