Received for publication: December 27, 2015; Accepted: January 14, 2016 Abstract: Phylogenetic study of the four sections (Aporum, Crumenata, Strongyle, and Bolbidium) of genus Dendrobium (family Orchidaceae) was conducted using molecular data. Classifications based on morphological characters have not being able to clearly divide these four sections neither do they supported their monophyly origin. Therefore, deeper and detailed analysis especially using molecular data is required to ascertain their status. Molecular evidences were used to clarify their relations either to lump them into one section or reduce them into two. The study has been carried out for the 34 species of Dendrobium using Maximum Parsimony (MP). Three nucleotide sequences data sets from two distinct genomes chloroplast DNA genes (rbcL and matK) and nuclear ribosomal DNA (ITS) were used to construct cladograms. The results that obtained from the Internal Transcribed Spacer (ITS) gene showed that the nuclear genes are reliable marker for the phylogenetic study of Dendrobium compared to chloroplast DNA with low resolution level among sections. Chloroplast DNA has been the most confusion in the species identification. Many extensively used source of data in the plant problems remain in the classification of this large phylogenetic analysis. The use of cpDNA has been genus based on morphological characters, so reviewed widely2. Some of the chloroplast genes advanced studies using molecular methods are commonly used in phylogenetic studies are needed. For respectively4,5. This genus was recognized by example, many species have laterally flattened Olofswartz in 1799 for the first time, as cited in leaves, some have terete leaves, and again many Seidenfaden and Wood (19924). Dendrobiums are have 'normal' (dorso-ventrally flattened) leaves. In distributed in the tropical and subtropical regions addition, some have stems with swollen in South, East and Southeast of Asia, north of internodes, while others have wiry stems. Many Australia, New Zealand and New Guinea6. They have thin-textured flowers that last only a single are one of the most popular orchids because of day, others (in section Aporum) have fleshier their medicinal and commercial values7. In flowers that last longer for more than two or three addition, genus Dendrobium is also famous due to weeks. Therefore, the main problems are difficulty their floriferous flower sprays, wide variety of in identifying species within the complex of the colors, sizes and shapes, year-round accessibility, four sections in the genus Dendrobium (Aporum, and long flowering life of several weeks to Strongyle, Crumenata and Bolbidium) through months8.
