MB 5547 Experimental Parasitology (1983-2005) Designed for students interested in basic research of parasitology, this course includes lectures, seminars and discussion with comprehensive coverage of the major tropical parasites currently under investigation. Students are to learn laboratory handling of Entamoeba, Leishmania, Plasmodium, Trypanosoma and other parasites as experimental models and their applications to resolve problems in tropical medicine and biology of parasitism in general. Spring quarter. Two hrs of lecture and three hrs of lab/wk. MB 5520 Molecular Parasitology (Since 1983 as needed) This course involves conceptual and practical aspects of applying new biotechnology to studying parasites and parasitic mechanisms of major tropical diseases. Emphasis is placed on biotechnology in handling proteins, DNA and RNA. Students are to actively participate in searching and discussing current literature as well as in project-oriented research. Research topics are catered to the background, training and/or interests of the students. Winter quarter. 3 hrs/wk lecture or discussion. Chang and faculty. 4 KPChang CV (Con’t) RESEARCH OBJECTIVES AND CURRENT PROGRAMS: Our current supports and emphasis are to develop photo-inactivated Leishmania genetically and chemically as non-viable, but delivery competent carrier for immuno-therapy and -prophylaxis against leishmaniasis and other infectious/malignant diseases. My main research interest is to study molecular structures and biological functions of extrachromosomal self-replicating entities in eukaryotic cells. Biological models studied include parasitic protozoa in mammalian cells, endosymbiotic bacteria in insects and protozoa, and extrachromosomal circular DNA molecules amplified from chromosomes in response to drug selection. Emphasis is placed on the understanding of molecular integration and discordance of intracellular interactions. Experimental approach is multidisciplinary. My work supported by NIH has been focused on the Leishmania model for molecular mechanisms of microbial virulence. Specific infection-related molecules are invasins/evasins (invasive/evasive determinants) of the parasites, e. g. zinc protease/major surface glycoprotein (gp63), microsomal glycosyltransferase in the dolichol pathway of protein glycosylation and the ATP scavenging enzyme, e. g. nucleoside diphosphate kinases. Gp63 expression is regulated posttranslationally by N-glycosylation, which is in turn controlled by the glycosyltransferase gene expression. Secretory nucleoside diphosphate kinase is under study as an evasive determinant to prevent ATP-induced cytolysis and P2X7-mediated apoptosis of macrophages. My past and current research includes the following specific topics: (1) Molecular Biology: Structure, regulation and expression of virulence and virulence-regulating genes cloned from Leishmania DNA studied by molecular genetic approach, i. e. gene replacement and transfection; (2) Biochemistry: Purification and characterization of Leishmania gp63, microsomal N-acetylglucosamine-1-phosphate transferase and the ATP scavenging enzymes; (3) Cell Biology: Post-translational regulation of these molecules and their activities in intracellular parasitism of macrophages by Leishmania; and (4) Molecular immunology: Genetic analysis of immunologically active parasite epitopes as the pathogenic determinants responsible for the clinical symptoms (=virulence) manifested in human leishmaniasis. Prophylactic and diagnostic potentials of these epitopes targeted for preparing attenuated mutants as vaccines and recombinant products as antigens for diagnosis, prophylaxis and therapy; (5) Molecular epidemiology: Genetic/genomic polymorphism of Leishmania single-copy/multicopy genes in relation to their evolution and virulence in clinico-epidemiology.
