No BSA or other carrier proteins that could interfere with conjugation. DATA Intracellular Staining by Flow Cytometry Detection of ADAMTS13 in HUVEC Human Cells by Flow Cytometry. HUVEC human umbilical vein endothelial cells were stained with Mouse Anti­Human ADAMTS13 Monoclonal Antibody (Catalog # MAB4245, filled histogram) or isotype control antibody (Catalog # MAB002, open histogram), followed by Phycoerythrin­conjugated Anti­ Mouse IgG F(ab')2 Secondary Antibody (Catalog # F0102B). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with saponin. PREPARATION AND STORAGE Reconstitution Reconstitute at 0.5 mg/mL in sterile PBS. BACKGROUND ADAMTS13 (a disintegrin and metalloproteinase with thrombospondin motifs 13) is a secreted multi­domain zinc protease primarily produced by the liver. In the circulation, it is responsible for cleaving von Willebrand factor (vWF) between Tyr842 and Met843. Defects in ADAMTS13 activity result in the generation of ultra large vWF, leading to thrombotic thrombocytopenic purpura (TTP).
