BACKGROUND Dengue is a major public health problem, affecting all tropical and many subtropical countries. It has been reported more than 200 thousand cases per year only in Americas (1). Sixty nine percent (69 %) of anti-infective drugs developed between 1981 and 2010 had been derived from or inspired by natural products, highlighting the importance of the natural resources in the research and development of new antiviral drugs (2). OBJECTIVE In this sense, this study aimed to evaluate the cytotoxicity and the in vitro anti-dengue activity of an Ethanolic Extract of Green Propolis (EEGP) (3) and an Exopolysaccharide extracted from liquid culture of Lactobacillus plantarum (ELP) (ATCC 8014). METHODS Cytotoxicity of EEGP and ELP was evaluated in C6/36 cells by MTT assay. Anti-dengue activity (DENV-2 American strain) of two different non-cytotoxic concentrations of EEGP (15 and 7.5 µg/mL) and ELP (250 and 125 µg/mL), added before or after virus inoculation, was evaluated by tittering cell culture supernatants by quantitative PCR at 48 and 72 h post infection. RESULTS AND CONCLUSION As results and conclusion, the 50% cytotoxicity (CC50) obtained values for EEGP and ELP were > 15 µg/mL and > 250 µg/mL, respectively. Although no anti-DENV-2 activity was found for EEGP or ELP, at the tested concentrations in C6/36 cell model, ongoing studies are in course in order to assess their potential anti-dengue activity on mammalian cell lines, as well as their indirect antiviral effects by immunomodulatory mechanisms.
