<p>Part 1: Modeling Mitosis You will investigate mitosis using a magnetic bead set.</p><p>1. Organize your magnetic bead set to represent interphase, prophase, metaphase, anaphase and telophase. 2. Draw each representation in the circles below or attach pictures of each into your lab notebook. 3. Label all parts visible in the cell for each phase as well as the name of each phase. 4. Answer Analysis Questions.</p><p>Part 2: Effects of Environment on Mitosis Scientists reported that a fungal pathogen, may negatively affect the growth of soybeans (Glycine max). Soybean growth decreased during three years of high rainfall, and the soybean roots were poorly developed. Close relatives of R. Anaerobis are plant pathogens and grow in the soil. A lectin-like protein was found in the soil around the soybean roots. This protein may have been secreted by the fungus. Lectins induce mitosis in some root apical meristem tissues. In many instances, rapid cell divisions weaken plant tissues.</p><p>You have been asked to investigate whether caffeine affects the number of cells undergoing mitosis in a different plant, using root tips.</p><p>You will be provided with untreated and caffeine-exposed roots. </p><p>Preparation Chromosome Squashes 1. Obtain a untreated onion root tip. 2. Place the onion root tip in 12 M HCl for 4 minutes using forceps. 3. Transfer the onion root tip to Carnoy’s fixative for 4 minutes. 4. Remove the slide from Coplin Jar containing 70% ethanol, dry with a scientific cleaning wipe, and label it treated/untreated and group initials. 5. Place the tip on the slide and cut off the distal (furthest away from center point) 2mm portion of the tip; discard the portion you cut off. 6. Cover the root tip piece with carbol-fuschin stain for 2 minutes. 7. Blot off the excess stain and cover the tip with 1-2 drops of H20 8. Place the cover slip over the tip and cover the cover slip with a scientific cleaning wipe. 9. Firmly press down on the coverslip with the eraser end of a pencil. Do not twist the slide, and be careful not to break the coverslip/slide. 10. Repeat for the treated onion root tip. Counting Cells and Analyzing Data 1. Observe the cells at high magnification (400x) 2. Look for well-stained, distinct cells 3. Within the field of view, count the cells in interphase and the cells in mitosis. Record your data in the correct data table. 4. Obtain another group’s tip of the same kind, count and record. Repeat on a 3rd tip. 5. Repeat for the other treatment 3x. 6. Cancer can occur when cells lost control of their cell cycle and divide abnormally. This happens when tumor-suppressor genes, such as p53 or Rb (retinoblastoma) are mutated. \</p><p>Case Study: HeLa Cells</p><p>HeLa cells are cervical cancer cells isolated from woman named Henrietta Lacks. Her cells have been cultured since 1951 and used in numerous scientific experiments. Henrietta Lacks died from her cancer not long after her cells were isolated. Answer the analysis questions.</p><p>Part 4: Modeling Meiosis</p><p>Meiosis resembles mitosis, but serves a very different purpose. Meiosis is a cell division resulting in the halving, or reduction, of chromosome number in each cell. A diploid organism has two sets of chromosomes (2n), while a haploid cell or organism has one set (1n). Meiosis produces gametes (ova and sperm) in animals and spores in fungi, plants and protists. Three other characteristics of meiosis are the exchange of genetic material (crossing over) between homologous chromosomes, the independent assortment of the chromosomes, and the separation of the alleles of the same gene. These characteristics, along with random fertilization, increase the genetic variability in the offspring. These mechanisms are essential to our understanding of genetics and evolution in sexually reproducing organisms.</p><p>1. Using the magnetic bead set from part 1 mitosis, model meiosis. 2. Part 5: Meiosis and Crossing over in Sordaria</p><p>The fungus Sordaria fimicola exchanges genetic material when two mycelia meet and fuse. The resulting zygote undergoes meiosis to produce asci, each ascus contains eight haploid spores. A single gene determines the spore color. A cross was made between wild type (+, black) and tan (tn) strains. The resulting zygote produces either parental asci, which have four black and four tan spores in a row (4:4 pattern) or if crossing over occurs, the ascus will have a 2:4:2, or a 2:2:2:2 arrangement. 1. Place a drop of water onto the microscope slide. 2. Gently scrape some perithecia from the agar plate near where the two strains meet. 3. Place a cover slip over the perithecia and put a scientific cleaning wipe over the cover slip. 4. Gently press down on the cover slip using the eraser end of a pencil. 5. Focus on high power (400x). If you do not see full ascus, please see your teacher for an alternative method of counting the ascus. 6. Count at least 50 asci, and score them as either parental or recombinant (crossing over). 7. Enter your data into the data table and make the calculations. One map unit equals the recombinant per 100 total events. The percentage of asci showing crossover divided by 2 equals the map units in this activity.
