<p>Version 1.0 Modified 6/6/2018</p><p>A super -Quick Guide to 1-D Gel Quantitation With Image Analysis 3.0</p><p>To successfully perform quantitative analysis on gels, MTP’s or membranes you’ll need to be comfortable with all the basic functions of gel analysis (steps 1-3 below). If you are comfortable with performing band sizing, then this will be a snap.</p><p>Step 1 – Gray Scale Adjustment</p><p>Step 2 –Color Separation </p><p>Step 3 – Lanes and Bands Definition</p><p>Step 4 – Quantification Setting and Data Generation Objective: Here you need to assign bands with a molecular weight (as opposed to a base pair designation). Instead of relating the migration distance to a bp length we are comparing an optical density to mass. We (the software) will make a regression between IOD and mass, provide a standard curve and estimate the mass of unknown bands.</p><p>1. To access the tools you’ll need set standards use either the 1-D Gel pull-down menu and select “Quantification Settings” or use the “Quantification Settings” icon (not sure what the icon is but it’s the one on the right of the “Open BP Curve” icon).</p><p>2. Once open you’ll need to set 4 parameters a. The channel of the marker b. The lane that has the maker c. The type of curve to use d. The plot type</p><p>3. Set the mass of each band by high lighting a band and then entering the mass in the lower left hand box. Click “Enter” to load the mass of the next band. </p><p>4. Check the box for each band you want included in the marker set and click “OK”</p><p>5. Click the “Volume Calculation” button (the calculator icon) to crunch the numbers.</p><p>6. To view the standard curve, go to the 1-D Gel pull-down menu and select “Open Standard Curve”.</p><p>NOTES: 1. The curve cannot be exported in this release. If you want to get the graphic out, do a screen capture and paste it wherever.</p><p>2. There is no “Template” function for this analysis.</p>
