Laboratory Safety Plan for: von Willebrand Factor (vWF) Metalloproteinase Assay Lab location Investigator: Primary User:

*Process vWF metalloproteinase elisa

*Hazardous Chemical/ Toxic chemicals barium chloride (poison) and o-phenylenediamine Chemical Class (carcinogen). Corrosive chemical H2O2. *Hazardous Equipment Centrifuge

*Potential Hazards Harmful is swallowed. Causes eye and skin irritation. Causes severe respiratory tract irritation. May cause severe digestive tract irritation with abdominal pain, nausea, vomiting and diarrhea. Target organs are kidneys, respiratory system, muscles, blood and cardiovascular systems. Hydroxide peroxide causes severe skin irritation and burns with prolonged contact with the skin. *Personal Protective Equipment Nitrile gloves, N100 respirator when weighing out chemicals, lab coat and protective eyeglasses or chemical safety goggles. *Engineering and Ventilation Minimize dust generation accumulation. Used in a well-ventilated area. Wet Controls wipe the balance stage and areas where powder has been used Designated Use Area for Solutions of chemicals are used only in the fume hood. Carcinogens, Mutagens, Reproductive or Acute Toxins Special Use Procedures Barium chloride is used for digestion of endogenous vWF and o-

phenylenediamine and H2O2 are used in the chromogenic solution for color reaction. Special Handling and Storage Wash thoroughly after handling. Wash hands before eating. Use with Requirements adequate ventilation. Store in a cool, dry, well-ventilated area away from incompatible substances. *Spill and Accident Procedures Clean up spills immediately. Place chemicals in a suitable disposal container.

*Waste Minimization Plan Barium chloride solution volume is kept to a minimum and made up as needed. O-phenylenediamine is weighed for each used. Amount is kept to a minimum. Hydrogen peroxide is used directly from bottle. *Hazardous Waste Disposal All unused o-phenylenediamine chromogenic solution will be discarded through the ECU hazardous waste management system. Less than .5ml of barium chloride is used per assay. Chemical is precipitated out of sample and supernatant used in assay. Decontamination Procedures Not applicable. Animal Care Precautions Not applicable. *Chemical Procurement The minimum amount of each chemical is kept on hand. Obtained when needed. *Revision Date Insert of Date: 3-5-2002
