Roberts Professor of Food Science Head of the Department of Food Science Approved by: Dr. ii ABSTRACT Numerous studies have indicated that the wine microbiome could generate various volatile compounds which could lead to distinguishing and different wine characteristics. However, little research regarding the wine microbiome is investigating specific microorganisms and their role within the entire microbial community under a more comprehensive sampling method. Thus, in this study we conducted direct sampling from Central and Northeast PA wineries instead of using lab-scale production to study the effect of the wine microbiome on the wine metabolome. Resulting grape and wine samples were characterized by next-generation sequencing and headspace-solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS). Collectively, the innovative sampling and experimental techniques provided a high-resolution picture of microbial dynamics and the resulting wine volatile profiles. Overall, we illustrated how microbial diversity and relative abundance of specific microorganisms change as fermentation progressed. Also, various wine volatile metabolites that are formed during the different fermentation stages were identified.

Chambourcin is the most abundant hybrid grape grown in Pennsylvania and is more resistant to cold temperatures and fungal diseases compared to Vitis vinifera. Here, non-Saccharomyces yeasts were isolated from spontaneously fermenting Chambourcin must from three regional vineyards. Using cultured-based methods and ITS sequencing, Hanseniaspora and Pichia spp. were the most dominant genus out of 29 fungal species identified. Five strains of Hanseniaspora uvarum, H. opuntiae, Pichia kluyveri, P. kudriavzevii, and Aureobasidium pullulans were characterized for the ability to tolerate sulfite and ethanol. Hanseniaspora opuntiae PSWCC64 and P. kudriavzevii PSWCC102 can tolerate 8–10% ethanol and were able to utilize 60–80% sugars during fermentation. Laboratory scale fermentations of candidate strain into sterile Chambourcin juice allowed for analyzing compounds associated with wine flavor. Nine nonvolatile compounds were conserved in inoculated fermentations. In contrast, Hanseniaspora strains PSWCC64 and PSWCC70 were positively correlated with 2-heptanol and ionone associated to fruity and floral odor and P. kudriazevii PSWCC102 was positively correlated with a Citation: Feng, C.T.; Du, X.; Wee, J. Nestled among the gently rolling hills of Chautauqua County lie six lakes 'lorthernmost which are the very/l'lIKKIoHM""ll'ltCOPYRIGHT headwaters of the Ohio valley Drainage Basin. The largest of these is Chautauqua Lake, from which the County and Chautauqua Institution derive the ir names. Our ChautauquaCHAUTAUQUALake muskallunge are called ''Muskallunge Ohiens is". Cassadaga and Bear Lake, in the Town of Stockton, are relatively smaller. Mud Lake and East Mud Lake in the Town of Villenova are the sources of the Conewango River, and COUNTY Findley Lake in the western edge of the County flows into the west branch of French Creek and finally into the Allegheny at Franklin,HISTORICALbelow Oil City. The highest of these lakes is Findley Lake on our western border with a Sea Level Elevation of 1420; West Mud Lake and East Mud Lake are 1369' and 1342' respectively; Chautauqua 1308; SOCIETY and Cassadaga and Bear Lake about 1303'. After all, over 90% of wine produced in this country comes from the Golden State. Famous wine regions like Napa Valley, Sonoma and Santa Barbara County are wildly popular for travelers, but a number of up-and- coming California wine regions are producing stellar grapes – and wines worth seeking out. Below are some of the up and comers that should be on your California wine radar. Lake County Bordered by the famous wine regions of Napa (and Mt. St. Helena) directly south, Sonoma to the southwest and Mendocino to the west, Lake County today produces quite a few fine wines in its own right. Carved out of Napa County in 1861, the area combines a few small towns, a hardy landscape, relatively high elevations, pear orchards and a multitude of vineyards. Visitors will be pleased to find high-quality wines (from approximately 35 wineries) without crowds, traffic or high prices. Vintners in Lake County have learned that their volcanic soils, hot days and mild nights, elevation, cool breezes from the Pacific and a strong will to produce high-quality wines are helping their region prosper. Big wines such as Cabernet Sauvignon, Syrah and Petite Sirah do well here as well as delicious Sauvignon Blancs.

February 11, 1882 COUNTY The steamboat Cincinnati being built for Burroughs Bros. at the Boatlanding is nearing completion.HISTORICALIt will be launched the first of next month. Its owners claim it will be the staunchest and speediest on the lake. SOCIETY April 8, 1882 WESTFIELD, The launch of the Cincinnati is announced for this after- noon. She is owned by Burroughs Bros. of Jamestown. She will have as officers this s1l1llllrer: E. A. Burroughs, Captain, C. W. Burroughs,NY Engineer, 2012 Everett Burroughs. Clerk, and Porter Payne, Pilot. She is 132 Foot long, 26 foot beam, and has a propeller wheel 5 foot 6 inches in diameter. There will be two decks, the ladies cabin on the second deck. She will be driven by a 400 horsepower engine and boiler. The machinery is promised by the 15th of April and she will be ready for business the first of May. The keel was laid on the 20th of OctOber. The entire work was under the charge - 2 - of Capt. J. W. Pierce of Evansville, Indiana, who promises the steamboat will be in eveI7/ way to be equal to a:rry steamboat on the lake. April 8! 1882 The Cincinnati commenced to move down the ways at 3:45 P.M.

    The Alden Lane Nursery Grape Collection Autumn Royal Seedless Large, seedless dark purple to black grape. Vigorous vine that responds best to spur pruning. Appeal rests on large berry size and late maturity. Ripens: Late September to Mid-October Black Monukka Seedless A medium size black seedless grape with a crisp sweet flavor. This is a hardy European variety which can take temperatures as low as 10 °F. A good fresh grape or suitable for raisins. Ripens: August into September Cane or Spur pruning Cabernet Sauvignon Small, round, purple-black grape with many seeds that make a distinctive world-renown red wine. Excellent producer in our area. Ripens late September to October Champagne (Black Corinth or Zante Currants) Although not a true currant, when dried they are referred to as Zante currants and used extensively in baking. It is an old Greek variety. Grape is dark red to black, firm skinned juicy and crunchy with a sweet spicy flavor when ripe. It is seedless. Ripens mid August. Cane or Spur prune Chardonnay For white burgundy style wines. Small round berry. Vigorous vine. Very cold hardy. Ripens: Late August to September Cane pruning Concord Seedless A medium size black seedless version of the classic Concord grape with all its hardiness, vigor, disease resistance and flavor. Excellent for table, juice and jams. Ripens: August Cane or Spur pruning Crimson Seedless Large red seedless table grape. Medium sized clusters of firm, crisp fruit with a sweet flavor. Holds well on vine after ripening. Ripens: October Cane pruning Delight Large. Dark greenish yellow, resembling Thompson Seedless in color.
