Figure S1. Fluorescein-labeled SH3 domain displayed similar affinities for their

peptide targets as the wild-type proteins. A peptide, YEDTPPPLPPKPKFR, derived from HPK1 and shown to bind to both the PLCγ1 SH3 and the Grb2 SH3N domain was

chemically synthesized and labeled at the N-terminus with biotin. The biotinylated

peptide was mixed with either the wt or fluorescein-labeled SH3 domain in solution, and the resulting peptide-SH3 complex was pulled down by streptavidin-agarose beads and

resolved on SDS-PAGE. The figure shows that fluorescein-labeling (via a C-terminal

Gly-Gly-Cys triad) does not affect binding of an SH3 domain to its target peptide. f-SH3, fluorescein-labeled SH3; Bt-peptide, biotinylated peptide.
