Mueller, III Director Federal Bureau of Investigation Before the Committee on Homeland Security and Governmental Affairs United States Senate At a Hearing Entitled “Homeland Threats and Agency Responses” Presented September 19, 2012 Good morning, Chairman Lieberman, Ranking Member Collins, and Members of the Committee. Thank you for the opportunity to appear before the Committee today and for your continued support of the men and women of the FBI. As you know, the Bureau has undergone unprecedented transformation in recent years. Since the attacks of September 11th, we have refocused our efforts to address and prevent emerging terrorist threats. The terrorist threat is more diverse than it was 11 years ago, but today, we in the FBI are better prepared to meet that threat. We also face increasingly complex threats to our nation’s cyber security. Nation-state actors, sophisticated organized crime groups, and hackers for hire are stealing trade secrets and valuable research from America’s companies, universities, and government agencies. Cyber threats also pose a significant risk to our nation’s critical infrastructure. As these threats continue to evolve, so too must the FBI change to counter those threats. We must continue to build partnerships with our law enforcement and private sector partners, as well as the communities we serve. Above all, we must remain firmly committed to carrying out our mission while protecting the civil rights and civil liberties of the people we serve. -

Status Update and Life History Studies on the Regal Fritillary

W?gner et al, Introduction With its fiery orange forewings, iridescent blue-black hindwings, and 10-centimeter (cm) wingspan, the regal fritillary was one of Ne\; England's most magnificent grassland insects. Although always locali' distributed, it formerly occurred in all six New England states (Scud Status Update and Life History Studies 1889, Denton 1900, Klots 1951, Opler 1983, 1992). Preferred hahita included marshes and swamp edges, wet meadows, fields, pastures. on the Regal Fritillary (Lepidoptera: and native grasslands (Denton 1900, Weed 1926, Clark 1932, Clark a" Nymphalidae) Clark 1951, Kiots 1951, Schweitzer 1992, Glassberg 1993; see also 1). An examination of the numbers of specimens in collections and accounts in the literature indicates that the species reached its greatc. - abundance in New England in Massachusetts on the sandplain grass- David L. Wagner, Matthew S. Wallace, lands of Martha's Vineyard, Nantucket Island, and other offshore George H Boettner, and Joseph S. Elkinton islands (Scudder 1889, Jones and Kimball 1943). On the mainland an: on Block Island, RI, the species was commonly associated with wet meadows and human-maintained grasslands created by mowing, grazing, and other agricultural practices. As in other Speyeria. the larvae are violet specialists. New England's regal fritillary colonies were associated with ovate-leaved violet (V/olafimbriatula), lance- leaved violet (V lanceolata), common blue violet (V papllionacen and birdfoot violet (V pedata) (Schweitzer 1987, Cassie et al. in pre Abstract D. Schweitzer pers. comm.). Eastern populations of the regal fritillary (Speyeria idalia) have been declining since Adult males begin emerging in mid- to late June, followed one to at least the late 1940s. -

Control of Enzyme Activity using Chemical Rescue of Protein Structure by Yang Du A thesis submitted to Johns Hopkins University in conformity with the requirements for the degree of Master of Science in Engineering Baltimore, Maryland May 2016 ii Abstract A cell can modulate the level of protein activity by the regulation of the amount of protein. On the other hand, strategies for controlling of cellular protein activity may lie in regulating the protein’s specific activity directly. Based on this idea, we demonstrated a method for designing an effector site directly into the catalytic domain of an enzyme and using a chemical compound to regulate the protein’s activity. This approach is different from the traditional chemical rescue of enzymes because it relies on disruption and restoration of protein structure instead of disrupting the active site and having the chemical compound restore the chemical functionality to the active site as a means to achieve modulated function. In detail, we demonstrate the activity of TEM-1 can be disrupted by removing buried tryptophan or leucine side chain that serve as a buttress supporting the protein structure. Three cases of site-direct mutations (W227G & W286G, L247G & W286G, and L282G & W286G) to TEM-1 β-lactamase enzyme (BLA) were made. In all cases, we observe disruption of enzyme function. To test the theory of chemical rescue of protein structure, we then assay β-lactamase activity in the presence of specific chemical compounds designed to bind in place of the iii side chains of the mutated amino acids. We observed an 8- fold restoration of W227G/W286G enzyme function in the presence of 2- (1-naphthylmethyl)-isoquinoline (NMIQ) and 3 -methyl-2- (1-naphthylmethyl)-1,3-benzothiazol-3- ium (MNMBT). -

(2018) Abstract: The increasing emissions of carbon dioxide, methane and carbon oxide (collectively referred as C1 compounds) are likely to configure a major contribution to global warming and other envi- ronmental issues. The implementation of carbon capture and storage (CCS) is considered a crucial strategy to prevent global warming, but the overall costs of currently available CCS technologies are still prohibitive for its large-scale deployment. Using microorganisms capable of assimilating C1 com- pounds for producing value-added products could be an important driver for mitigating emissions and minimizing their deleterious consequences, while simultaneously deriving additional economic benefits from these compounds. This review summarizes the main microorganisms and metabolic routes being investigated, with special focus on both the products targeted and the current industrial initiatives. Based on the dynamic capabilities theory, the main goal of this paper is to address this research gap and to explore the rela- tionships between both internal and external knowledge accumulation, and ordinary organizational capabilities. This research also contributes to analyzing the complex effect of the family firm essence, influenced by both family involvement and generational involvement levels, as an antecedent of internal and external knowledge accumulation. Our analysis of 102 non-listed Spanish family firms shows that the family firm essence, which is influenced by the family involvement, strengthens only the internal knowledge accumulation but not the external one. Lignocellulosic biomass can be gasified to produce syngas, which is a gas mixture consisting mainly of H2, CO, and CO2. The major challenge of biomass gasification is the syngas’s final quality. Consequently, the development of effective syngas clean-up technologies has gained increased interest in recent years. Furthermore, the bioconversion of syngas components has been intensively studied using acetogenic bacteria and their Wood–Ljungdahl pathway to produce, among others, acetate, ethanol, butyrate, butanol, caproate, hexanol, 2,3-butanediol, and lactate. Nowadays, syngas fermentation appears to be a promising alternative for producing commodity chemicals in comparison to fossil-based processes. -

These constructs provide insight on the phenome of schizophrenia and show how pathway-phenotypes mediate the effects of genome X environmentome interactions on the symptomatology/phenomenology of schizophrenia.
