Oleo Sci. 62, (6) 391-401 (2013) Structure-activity Relationships for Bergenin Analogues as β-Secretase (BACE1) Inhibitors Yusei Kashima and Mitsuo Miyazawa* Department of Applied Chemistry, Faculty of Science and Engineering, Kinki University (3-4-1, Kowakae, Higashiosaka-shi, Osaka 577-8502, JAPAN) Abstract: Here we evaluated the inhibitory effects of bergenin analogues (2-10), prepared from naturally occurring bergenin, (1) on β-secretase (BACE1) activity. All the bergenin analogues that were analyzed inhibited BACE1 in a dose-dependent manner. 11-O-protocatechuoylbergenin (5) was the most potent inhibitor, with an IC50 value of 0.6 ± 0.07 mM. The other bergenin analogues, in particular, 11-O-3′,4′- dimethoxybenzoyl)-bergenin (6), 11-O-vanilloylbergenin (7), and 11-O-isovanilloylbergenin (8), inhibited BACE1 activity with IC50 values of <10.0 mM. BACE1 inhibitory activity was influenced by the substituents of the benzoic acid moiety. To the best of our knowledge, this is the first report on the structure-activity relationships (SAR) in the BACE1 inhibitory activities of bergenin analogues. These bergenin analogues may be useful in studying the mechanisms of Alzheimer’s disease. En- dothelial cell proliferation has been used as an in vitro model of angiogenesis. Trifolium pratense was suggested for cancer treatment in traditional medicine and scientific report. The present study was designed to investigate the effect of T. pratense hydroalcoholic extract on the human umbilical vein endothelial cell (HUVEC) apoptosis and nitric oxide (NO) secretion in vitro. Materials and Methods: Hydroalcoholic extract of T. pratense was prepared and its different concentrations (0, 12.5, 25, 50, 100, 200, 400, 800 μg/ml) were used for cell treatment. Cell viability was assessed by trypan blue, MTT and lactate dehydrogenase methods. Furthermore, nitric oxide (NO) secretion of the cells was measured by Griess reaction, and their apoptosis was evaluated by fluorescent dyes staining. One-way ANOVA was used for data analysis. Results: After 24, 48 and 72 hours treatments, the IC50 values were 1069.35, 174.70 and 68.07 μg/ml, respectively. T. pratense extracts exerted a significant difference between the groups treated compared to the control group (p<0.05) and increasing the dose significantly decreased cell via- bility. T. pratense extract significantly increased LDH in dose- and time-dependent manners in the cell culture medium (p<0.05).

The hydroxybenzoic acids are related to salicylic acid and salicin, the first compounds isolated that have a pharmacological activity. In this review we examine how a number of hydroxyphenolics have the potential to ameliorate cardiovascular problems related to aging such as hypertension, atherosclerosis and dyslipidemia. The compounds focused upon include 2,3-dihydroxybenzoic acid (Pyrocatechuic acid), 2,5-dihydroxybenzoic acid (Gentisic acid), 3,4-dihydroxybenzoic acid (Protocatechuic acid), 3,5-dihydroxybenzoic acid (α-Resorcylic acid) and 3-monohydroxybenzoic acid. The latter two compounds activate the hydroxycarboxylic acid receptors with a consequence there is a reduction in adipocyte lipolysis with potential improvements of blood lipid profiles. Several of the other compounds can activate the Nrf2 signaling pathway that increases the expression of antioxidant enzymes, thereby decreasing oxidative stress and associated problems such as endothelial dysfunction that leads to hypertension as well as decreasing generalized inflammation that can lead to problems such as atherosclerosis. It has been known for many years that increased consumption of fruits and vegetables promotes health. We are beginning to understand how specific phytochemicals are responsible for such therapeutic effects. Thus generation of lipoxygenase enzyme inhibitors have suggested being valuable. Individual variation was identified by the functional effects of Single Nucleotide Polymorphisms (SNPs). 696 SNPs were identified from the ALOX12 gene, out of which 73 were in the coding non-synonymous region, from which 8 were found to be damaging. In silico analysis was performed to determine naturally occurring flavonoids such as isoflavones having the basic 3- phenylchromen-4-one skeleton for the pharmacological activity, like Genistein, Diadzein, Irilone, Orobol and Pseudobaptigenin. O-methylated isoflavones such as Biochanin, Calycosin, Formononetin, Glycitein, Irigenin, 5-O-Methylgenistein, Pratensein, Prunetin, ψ-Tectorigenin, Retusin and Tectorigenine were also used for the study. Other natural products like Aesculetin, a coumarin derivative; flavones such as ajoene and baicalein were also used for the comparative study of these natural compounds along with acteoside and nordihydroguaiaretic acid (antioxidants) and active inhibitors like Diethylcarbamazine, Zileuton and Azelastine as standard for the computational analysis.

Van Heerden* Abstract Background: Malaria remains a signifcant public health challenge in regions of the world where it is endemic. An unprecedented decline in malaria incidences was recorded during the last decade due to the availability of efective control interventions, such as the deployment of artemisinin-based combination therapy and insecticide-treated nets. However, according to the World Health Organization, malaria is staging a comeback, in part due to the develop- ment of drug resistance. Therefore, there is an urgent need to discover new anti-malarial drugs. This article reviews the literature on natural products with antiplasmodial activity that was reported between 2010 and 2017. Methods: Relevant literature was sourced by searching the major scientifc databases, including Web of Science, ScienceDirect, Scopus, SciFinder, Pubmed, and Google Scholar, using appropriate keyword combinations. Results and Discussion: A total of 1524 compounds from 397 relevant references, assayed against at least one strain of Plasmodium, were reported in the period under review. Out of these, 39% were described as new natural products, and 29% of the compounds had IC50 3.0 µM against at least one strain of Plasmodium. Several of these compounds have the potential to be developed into≤ viable anti-malarial drugs. Also, some of these compounds could play a role in malaria eradication by targeting gametocytes. However, the research into natural products with potential for block- ing the transmission of malaria is still in its infancy stage and needs to be vigorously pursued.

Molecular docking studies were used to predict their binding affinity and interactions with amino acids residues from the active binding site of SARS-CoV-2 main protease, compared to boceprevir. Deep structural insights and quantum chemical reactivity analysis according to Koopmans’ theorem, as a result of density functional theory (DFT) computations, are reported. Additionally, drug-likeness assessment in terms of Lipinski’s and Weber’s rules for pharmaceutical candidates, is provided. The outcomes of docking and key molecular descriptors and properties were forward analyzed by the statistical approach of principal component analysis (PCA) to identify the degree of their correlation. The obtained results suggest two promising candidates for future drug development to fight against the coronavirus infection.
