Selection and Evaluation of a Silver Nanoparticle Imaging Agent for Dual-Energy Mammography Abstract Over the past decade, contrast-enhanced (CE) dual-energy (DE) x-ray breast imaging has emerged as an exciting, new modality to provide high quality anatomic and functional information of the breast. The combination of these data in a single imaging procedure represents a powerful tool for the detection and diagnosis of breast cancer. The most widely used implementation of CEDE imaging is k-edge imaging, whereby two x-ray spectra are placed on either side of the k-edge of the contrast material. Currently, CEDE imaging is performed with iodinated contrast agents. The lower energies used in clinical DE breast imaging systems compared to imaging systems for other organs suggest that an alternative material may be better suited. We developed an analytical model to compare the contrast of various elements in the periodic table. The model predicts that materials with atomic numbers from 42 to 52 should provide the best contrast in DE breast imaging while still providing high-quality anatomical images.

  • AP0599 Nanoparticle Decoration of Carbon Nanotubes by Sputtering
Hiden Reference: AP0599 Hiden Product: EQP 1000 Nanoparticle decoration of carbon nanotubes by sputtering Nanoparticle-decorated carbon nanotubes (CNTs) are effective chemical and biological sensors, surfaces for heterogeneous catalysis, photovoltaics, and conformal thermal interface materials for electronics. The particle morphology on the CNT sidewalls strongly affects the properties and performance of metal-nanotube hybrids for such applications. Often nanoparticles are deposited by electrochemical methods, which generally require time consuming treatments with strong acid for surface defect production, which can result in a compromise of the intrinsic mechanical or transport properties of the CNTs, inhibiting their multi-functionality. We have examined physical vapor deposition techniques as scalable alternatives to electrochemical treatment for in situ growth of metal nanoparticles on the sidewalls of multi-wall carbon nanotubes (MWCNTs). Vapor phase growth of gold, nickel and titanium metal nanoparticles on multi-wall carbon nanotube (MWCNT) bucky paper was investigated. The size and distribution of nanoparticles was dependent on the intrinsic binding energy of the elemental metals, where metals with larger cohesive energies exhibited a higher nanoparticle density and smaller particle diameters. Particle diameters for any metal could be altered to mimic that of metals with different binding energies by in situ modification of the MWCNT surfaces by energetic metal ions (characterized with a Hiden EQP 1000 as shown in Figure 1) during their growth, where removal of a carbon atom from a MWCNT surface requires incident ions kinetic energies > 5-7 eV. Control of the ariel density, diameter and morphology of metal nanoparticles grown on as-received and annealed multi- walled carbon nanotube sidewalls by sputtering was demonstrated for gold, nickel and titanium.

For the synthesis, 50 mL deionized water was brought to a boil and then 365 μL of 34 mM trisodium citrate dihydrate (Sigma Aldrich, St. Louis, MO, USA) and 211 μL AgNO3 (Sigma Aldrich) were added, followed by drop-wise addition of freshly prepared 250 μL NaBH4 (Sigma Aldrich). Upon addition of NaBH4, the solution immediately turned yellow, and the mixture was allowed to boil for 15 min before removing the nanoparticles from heat and allowing them to come to room temperature. Nanoparticles were purified with regenerated cellulose (MWCO 50,000) centrifugal filter units (EMD Millipore) where 15 mL of the nanoparticle suspension were centrifuged at 1500 g for 4 min and then resuspended in deionized water, with the centrifuge/resuspension steps repeated in triplicate.

Some additional studies were also performed: measurement of fullerenol nanoparticle size, zeta potential, and the influence of fullerenol on the ionizing radiation-induced damage to PMBCs. Fullerenol C60(OH)36 demonstrated an ability to accumulate in PBMCs. The accumulation of fullerenol in those cells did not have a significant effect on cell survival, nor on the distribution of phosphatidylserine in the plasma membrane. However, fullerenol-induced depolarization of the mitochondrial membrane proportional to the compound level in the medium was observed.

A nanometre is one billionth of a metre (0.000 000 001 m). So, it may not come as a surprise that nanoparticles of material show different properties compared to larger particles of the same material. The surface area to volume ratio for a material or substance made of nanoparticles has a significant effect on the properties of the material. Firstly, materials made up of nanoparticles have a relative larger surface area when compared to the same volume of material made up of bigger particles. r For example, let us consider a sphere of radius r The surface area of the sphere will be 4πr2 The volume of the sphere = 4/3πr3 Therefore the surface area to the volume ratio will be 4πr2/(4/3πr3) = 3/r It means that the surface area to volume ratio increases as the radius of the sphere decreases and vice versa. It also means that when a given volume of material is made up of smaller particles, the surface area of the material increases. Therefore, as particle size decreases, a greater proportion of the particles are found at the surface of the material. For example, a particle of size 3 nm has 50% of its particles on the surface; at 10 nm, 20% of its particles are on the surface; and at 30 nm, 5% of its particles are on the surface. Therefore, materials made of nanoparticles have a much greater surface area per unit volume ratio compared with the materials made up of bigger particles. This leads to nanoparticles being more chemically reactive.

  • Ultrasound-Mediated Delivery of RGD-Conjugated Nanobubbles
RSC Advances View Article Online PAPER View Journal | View Issue Ultrasound-mediated delivery of RGD-conjugated nanobubbles loaded with fingolimod and Cite this: RSC Adv., 2020, 10, 39348 superparamagnetic iron oxide nanoparticles: targeting hepatocellular carcinoma and enhancing magnetic resonance imaging† Xin-Min Guo, * Jia-Lin Chen, Bao-Hui Zeng, Ji-Chuang Lai, Cui-Yan Lin and Mei-Yan Lai Nanobubbles (NBs) are considered to be a new generation of ultrasound-responsive nanocarriers that can effectively target tumors, accurately release multi-drugs at desired locations, as well as simultaneously perform diagnosis and treatment. In this study, we designed theranostic NBs (FTY720@SPION/PFP/RGD- NBs) composed of RGD-modified liposomes as the shell, and perflenapent (PFP), superparamagnetic Creative Commons Attribution-NonCommercial 3.0 Unported Licence. iron oxide nanoparticles (SPION), and fingolimod (2-amino-2[2-(4-octylphenyl)ethyl]-1,3-propanediol, FTY720) encapsulated as the core. The prepared FTY720@SPION/PFP/RGD-NBs were black spheres with a diameter range of 160–220 nm, eligible for enhanced permeability and retention (EPR) effects. The calculated average drug loading efficiency (LE) and encapsulation efficiency (EE) of the FTY720@SPION/ PFP/RGD-NBs were 9.18 Æ 0.61% and 88.26 Æ 2.31%, respectively. With the promotion of low-intensity focused ultrasound (LIFU), the amount and the rate of FTY720 released from the prepared NB complex were enhanced when compared to the samples without LIFU treatment. In vitro magnetic resonance imaging (MRI) trials showed that the prepared FTY720@SPION/PFP/RGD-NBs had a high relaxation rate and MRI T2-weighted imaging (T2WI) scanning sensitivity conditions.

  • A Review on the Optimal Design of Magnetic Nanoparticle-Based T2 MRI Contrast Agents
magnetochemistry Review A Review on the Optimal Design of Magnetic Nanoparticle-Based T2 MRI Contrast Agents Nina Kostevšek Department for Nanostructured Materials, Jožef Stefan Institute, 1000 Ljubljana, Slovenia; [email protected] Received: 12 February 2020; Accepted: 25 February 2020; Published: 28 February 2020 Abstract: Relaxivity r2 and thus the contrast efficacy of superparamagnetic nanoparticles (NPs) can be enhanced via either NP’s magnetic properties or coating optimization. Numerous reports can be found about the investigation of the optimal iron oxide nanoparticles (IO NPs) size, shape, crystallinity and composition that yield high saturation magnetization (ms) values and, consequently, high r2 values. Although the use of an appropriate coating can boost up the NPs MRI contrast agent efficiency, this topic has been largely understudied. Therefore, in this review, the factors affording r2 enhancement of spherical magnetic NPs are discussed. Based on the literature, the requirements for an optimal surface coating that may increase r2 values and ensure stability and biocompatibility of NPs are listed. One of the best candidates that fulfil these requirements are liposomes with embedded magnetic NPs, so-called magneto-liposomes. The analysis of the literature elucidated the most appropriate phospholipid compositions for the relaxivity enhancement and for magneto-liposomes in vivo stability. Finally, the future directions in the development of NP-based contrast agents are given. For example, most of the synthetic NPs are recognized and eliminated as a foreign substance by the immune system. To overcome this issue, a design of a biomimetic, cell-membrane-based nanocarrier for contrast agents is proposed. Disguised with cell membranes, NPs or other active components can act as autogenous cells and thus ensure the inherent biocompatibility.
