Equipment:

Hot plate

Scale for measuring

Weigh boat

Thermometer

Buffer solution (add CyberSafe to buffer if using, 30 microliters/liter of buffer)

Protective mitt

0.8 g of Agarose Powder (1.2 g if using double well tray)

Flask

Gel trays

Combs for the gels Procedure

1. Gather all of your equipment.

2. Place the comb using the small 7 x 7 trays at the top of the tray (put in 2 combs if using tray with double wells)

3. Turn on the power of the hot plate. Be sure they are not placed close to the computer. Remember to be careful, they get very hot.

4. Measure 40 ml of buffer solution and pour into the flask (60 mL if using double tray). If you didn’t add CyberSafe to the buffer, you can add 5 microliters to the flask before boiling.

5. Measure .8 grams of Agarose Powder using a weigh boat. Remember to zero the weigh boat prior to measuring (1.2 g if using double well tray)

6. Add the agarose powder to the buffer solution

1 7. Heat the mixture until it boils (about one minute). BE CAREFUL HERE – IT CAN BOIL OVER!!! When it has boiled and the solution is clear, turn heat off and obtain the thermometer.

8. In a few minutes place the thermometer inside the solution. You want the solution to be cooled to 60 degrees C.

9. When the solution has cooled, pour into the trays.

10. Once cooled, remove the combs

11. Take out the trays and wrap with plastic wrap and refrigerate until ready to use.

12. Let warm to room temp for several hours before running.

If you are using the gels within 24-48 hours you should be good and not have to soak them in buffer but if you want to you can in the plastic Ziploc boxes. 10 microliters of syber to 1000 mls of buffer. Buffer is included in the set.
