Part I: Summary of general activities related to the disease

2. Production and distribution of diagnostic reagents

We submitted the mycoplasma free CCO cell line to the American Type Culture Collection (ATCC) in 2003. This and the type CCV isolate are available from ATCC. CCV DNA and infected cell lysates are available for positive controls for PCR and antigen based tests. Neutralizing monoclonal antibody is available. There were no requests for these reagents in 2011.

3. International harmonisation and standardisation of methods for diagnostic testing or the production and testing of vaccines

a) Establishment and maintenance of a network with other OIE Reference Laboratories designated for the same pathogen or disease and organisation of regular inter-laboratory proficiency testing to ensure comparability of results

b) Organisation of inter-laboratory proficiency testing with laboratories other than OIE Reference Laboratories for the same pathogens and diseases to ensure equivalence of results

4. Preparation and supply of international reference standards for diagnostic tests or vaccines

5. Research and development of new procedures for diagnosis and control

1. We are evaluating different extraction and enrichment procedures to enhance our ability to detect carrier fish.

2. We are evaluating the potential of controlling CCVD using vaccinations.

3. We have established a BAC system for producing recombinant CCV for vaccine work.

4. We have established a Microarray that contains 70 CCV genes on it for gene expression studies.

5. We are building and characterizing a CCV case isolate library to be used for evaluating diagnostic tests and genetic/antigenic diversity of CCV.

6. We have performed an initial evaluation of our case library and determined that there are at least 2 genogroups of the virus. We have established a PCR method that distinguishes the genogroups and are in the process of evaluating these tests on a larger number of samples.
