Institutional Animal Care and Use Committee (IACUC) Standard Operating Procedure

GUILLOTINE/SHARP SCISSORS USE

1. Purpose This SOP is intended to provide information so that researchers at ISU using rodent guillotines or sharp scissors will conform to the AVMA Guidelines for Euthanasia as well as to meet requirements for AAALAC’s triennial site visits. The purpose of this SOP is to assure that guillotines or sharp scissors are kept clean and in sharp working order to assure humane euthanasia and that this information is properly documented.

2. Scope This SOP is applicable to all ISU staff, research investigators, and technicians who use guillotines or sharp scissors for euthanasia.

3. General Requirements The principal investigator (PI) must ensure that all personnel using the guillotine or sharp scissors are properly trained and proficient in their use. The PI should maintain a training record in the lab where the guillotine or sharp scissors are located or used. Contact Laboratory Animal Resources (LAR) if assistance is needed.

4. Procedure 1. Prior to use, the guillotine or sharp scissors should be inspected for rust, smoothness of operation, and cleanliness. 2. A suggested method to ensure that guillotine or sharp scissors blades are sufficiently sharp is to use a fresh rodent carcass of an animal that has been euthanized in another IACUC-approved manner. 3. Personnel using the guillotine or sharp scissors must document usage either in a log or in a table provided. This log or table must be available to the IACUC for semiannual inspections. 4. After use, the guillotine or sharp scissors should be washed with water, disinfected, and given a final 70% alcohol rinse.

Office for Responsible Research Page 1 of 4 Iowa State University 5. As needed, the guillotine or sharp scissors should be lubricated with a silicone spray and/or sharpened. It is the PI’s responsibility to make sure the guillotine or sharp scissors are clean and sharp. 6. Record all instances of lubricating or sharpening of the guillotine or sharp scissors in the guillotine/scissors log (template below).

7. An alternative to guillotine or sharp scissors is a sterile, one-time-use scalpel blade. These will not be re-used, so there is no need to follow this SOP when using one- time-use scalpel blades. Dispose of scalpels properly in a puncture-resistant container.

5. Roles and Responsibilities This section is in a table format. Each individual participating in the duties outlined in the SOP (i.e., animal caretaker, veterinarian, animal researcher, etc.) will be listed in their own row within the “Roles” column. In the corresponding “Responsibilities” column, the tasks that group of individuals is responsible for should be described.
