Business Plan 3 Company

Defytime Ltd is headquartered in Auckland New Zealand, with labs co-located in The University of Auckland and research and development in Reno, Nevada USA. Our first anti-aging clinic is located in Vanuatu. Our goal is to reverse the human aging process and cure diseases linked to aging by activating telomerase and thus extending telomeres, leading the cells to return to a state of youthful gene expression and function. This cutting edge research is led by director Dr. Bill Andrews. Dr. Andrews has worked in the biotech industry for over 35 years, focusing the last 20 years on finding ways to extend human lifespan through the intervention of telomere shortening in human cells.

Business Plan 4 Our Team

Dr. Bill Andrews Laura A. Briggs, Ph.D. Founder & President Vice President of Research & Discovery Dr. Andrews is the founder and President of Sierra Science Laura Briggs received her B.S. degree in Nutrition in 1993 and LLC, a biotech company in Reno, Nevada focused exclusively her Ph.D. in Environmental Science and Health in 2000 from on finding drugs that will transiently induce the expression of the University of Nevada, Reno. After a one year post-doctoral endogenous telomerase in human cells. position at UNR she joined Sierra Sciences, known as Andrews Inc in 2001. In addition to coordinating research and development at Dr. Andrews earned his Ph.D. in Molecular and Population Genetics at Sierra Sciences, Dr. Briggs is also serving as the Biology Lab Coordinator the University of Georgia in 1981. He was a Senior Scientist at Armos Corp for Truckee Meadows Community College (TMCC) and has collaborated and Codon Corp, Director of at Codon and at Geron on research projects at the V.A. Medical Center in Reno, Nevada. Corp, and Director of Technology Development at EOS Biosciences. While Director of Molecular Biology at Geron Corp from 1992 to 1997, Lancer Brown, M.S. Dr. Andrews was one of the principal discoverers of both the RNA and protein components of human telomerase and was awarded 2nd place Program Director of Screening as "National Inventor of the Year" in 1997 for this work. Lancer Brown received his B.S. and M.S. degrees in He is named inventor on 45 US issued telomerase patents. Dr. Andrews in 2003 from the University of Nevada, Reno is highly respected and widely known for his expertise and passion and was one of three students in the inaugural advanced BS/ for curing aging and TEDS. He has been a featured expert in many MS Biotechnology Program. He distinguished himself as the documentaries and is a frequent guest speaker at prestigious events. first student to complete the program. Lancer came to Sierra Sciences as Dr. Andrews is also an ultra marathon runner whose ultimate goal is an intern while completing his degree. Following graduation, he joined to run a 7-minute mile at the age of 130. Sierra Sciences full-time where he has proven to have remarkable ability to engineer genes and DNA. He has recently been promoted to Program Director of Screening.

Business Plan 5 Cell Telomeres Chromosome Telomere

Telomeres are an essential part of human cells that affect how our cells age. plays a critical role in normal growth, maintenance and DNA repair of all human tissues, including skin.

Telomeres are repeated sequences of DNA at the ends of each Telomere chromosome and are key genetic elements involved with the regulation of cell division. Telomeres shorten every time a cell divides, and once telomeres reach a critically short length, the cell either dies by apoptosis or stops dividing and senesces. Telomeres are the caps at the end of each strand of DNA that protect our chromosomes, like the plastic tips at the end of shoelaces. Without the coating, shoelaces become frayed until they can no longer do their job, just as without telomeres, DNA strands become damaged and our cells can’t do their job. Telomeres are shortened as we age, but telomeres can also be shortened by stress, smoking, obesity, lack of exercise and a poor diet.

As cells divide over time telomeres shorten, and eventually cell division stops.

Business Plan 6 Telomerase

Telomerase is a naturally occurring enzyme that maintains telomeres and prevents them from Telomerase shortening during cell division. Telomerase, also called telomere terminal transferase, is a ribonucleoprotein that adds the polynucleotide “TTAGGG” to the 3’end of telomeres, at the ends of eukaryotic chromosomes. U C Telomerase is a reverse transcriptase enzyme that carries its own RNA RNA template C C A A molecule (with the pattern of ”CCCAAUCCC” in vertebrates), which is G T A used as a template for adding new bases onto the ends of telomeres. Business Plan 7 What is Aging?

Ageing (British English) or aging (American English) is just the process of becoming older. The term is used to refer to biological aging of human beings, animals and other organisms, but can also refer to individual cells within an organism (cellular aging) or to the population of a species (population aging). In humans, aging is an accumulation of changes over time, encompassing physical, psychological, and social change. Reaction time may slow with age, while knowledge and wisdom may expand. Aging is one of the greatest contributing risk factors for most human diseases, and of the roughly 150,000 people who die each day across the globe, about two thirds die from age-related causes.

Business Plan 8 Causes & Treatment of Aging The activation of telomerase in aged mice rejuvenates the entire body

Telomerase Induction The causes of aging can be ascribed to damage, whereby the accumulation of externally induced damage (such as DNA point mutations) may cause biological systems to fail, or to programmed aging, whereby internal processes (such as DNA telomere shortening) may cause aging. Telomere-shortening can be seen as a programmed aging ‘disease’, and could be cured by lengthening telomeres via Telomerase Induction.

Defytime proposes 2 different Telomerase Induction methods: • Anti Aging Therapeutic used in creams, supplements, and drugs • Hybrid Vector Solution (injection of a virus), available in our clinic in With telomerase aging is reversed Without telomerase aging continues Vanuatu for VVIP patients. The next 2 pages explain how these processes work.

Business Plan 9 Repressor Anti Aging Therapeutic Protein no Telomerase production

Chromosome

Telomerase Induction Method 1 Regulator Site Telomerase Gene Telomerase can be naturally produced by your cells to repair telomeres, but this function is shut off by a repressor protein binding to a regulator site or ‘switch’ on the chromosome.

• Defytime introduces a therapeutic element that binds to the Repressor repressor protein. Protein Telomerase production activated • This prevents the repressor protein from binding to the regulator site, Anti-aging Therapeutic allowing telomerase production to be activated. Chromosome • The telomerase produced by your cells can then repair the portions of the telomeres lost in cell division. Regulator Site Telomerase Gene

Business Plan 10 Hybrid Vector Solution + = Cloning Activated Adeno-Associated Virus Human Telomerase Gene (AAV) Viral Vector Hybrid Vector Telomerase Induction Method 2 In Defytime Telomere Lengthening Therapy, a hybrid vector combining the activated human telomerase gene with an adeno associated virus (AAV) is made and cultured. • The adeno associated virus does not cause disease, and has a very mild immune response. • The AAV transfers the hybrid vector (section of DNA) into your cells, where it can produce telomerase. • The telomerase produced by your cells can then repair the portions of the telomeres lost in cell division. Transfers into cells and onto Hybrid vector solution genomic DNA to produce telomerase. injected into the bloodstream

Business Plan 11 TAM CO314818
